Characterization of a novel soluble c-type cytochrome in a moxD mutant of Methylobacterium extorquens AM1

Abstract: Methylobacterium extorquens AM1 contains a novel c-type cytochrome, called cytochrome c-553, previously thought to be a precursor of the electron acceptor (cytochrome cL) for methanol dehydrogenase. Its amino acid composition and serological characteristics show that it has no structural relationship to cytochrome cL. It usually comprises less than 5% of the total c-type cytochromes. In a moxD mutant, which contains neither methanol dehydrogenase nor cytochrome cL, it comprises 30% of the soluble cytochrome an… Show more

“…Data from the literature suggest that a cytochrome c may be involved in accepting electrons from amicyanin (20,26), but in M. extorquens AM1, no amicyanin-specific cytochrome has been identified. However, a cytochrome c of unknown function, cytochrome c-553, has been identified in M. extorquens AM1 (16) and is therefore a possible candidate for this role. We were interested in determining whether the genes of unknown function in the methylamine utilization cluster described here encode cytochrome c-553, which is approximately 23 kDa in molecular mass.…”

“…However, a moxD mutant of M. extorquens AM1, which contains no methanol dehydrogenase protein, forms large amounts of that c553-type cytochrome (15). Its molecular mass of 23 kDa and its ability to bind CO, equal to that of cytochrome C553; in P. denitrificans (CO binding not shown here), suggest similarity between these c-type cytochromes in both organisms.…”

“…The physiological function of cytochrome C553 in M. extorquens AM1 is unknown at the moment. In vitro experiments show that this cytochrome mediates electrons between the cytochromes CL and CH (15), but the physiological relevance of this phenomenon is unknown.…”

“…The method used was that of BeardmoreGray et al (1983). except for determination of kinetic constants for cytochrome cL, when the more rapid assay of Day & Anthony (1990) was used.…”
