Characterization of a new human monoclonal antibody directed against the Vel antigen

Abstract: Serological characterization of the new monoclonal anti-Vel SpG213Dc showed a heterogeneous level of expression of the Vel antigen on the different RBCs. Our results suggest that the mAb SpG213Dc can be reliably used as a blood grouping reagent, thus allowing the mass-scale phenotyping of blood donors to strengthen rare blood banks with Vel-negative RBC units.

“…In fact, generating Vel‐specific hybridoma cell lines based on mouse immunization is likely to be difficult due to the high homology between human and mouse SMIM1 extracellular domain . A Vel‐specific antibody‐producing human hybridoma cell line was recently described, which was also generated from an immunized individual …”

“…In addition, using M3F5S‐IgM dispenses the use of an additional secondary antibody step to induce agglutination, as current human anti‐Vel sera contain mainly IgG antibodies. M3F5S‐IgM can therefore be used in an automated blood group analyzer, such as the Olympus PK7300, in contrasts to IgGs such as the previously described anti‐Vel IgG SpG213Dc …”

“…The classical serological method is not acceptable as a screening strategy for Vel− blood donors due to the scarcely available anti-Vel purified from Vel− individuals, which also displays heterogeneous levels of reactivity (Ballif et al, 2013;Cvejic et al, 2013;Storry et al, 2013;Haer-Wigman et al, 2015). However, monoclonal anti-Vel has recently been developed, side stepping this limitation (Danger et al, 2016).…”
