Peanut is a leguminous crop with an indeterminate growth habit that will continuously flower during the entire reproductive development stage. Flowering redundancy adversely affects the yield and quality of peanut. In this study, eight flowering and five yield-related traits were collected and comprehensively evaluated from a diverse germplasm panel consisting of 232 peanut accessions, aiming to provide a theoretical basis for improving the flowering habit and yield for future peanut breeding efforts. As a result, large phenotypic diversity was observed in 13 traits. Most of the traits suggested high heritability, except high effective flowering duration (HEFD), days to 100% flower cessation (DTC100), and yield per plant (YPP). Days to 90% flower cessation (DTC90), days to DTC100, flowering duration (FD), and low effective flowering duration (LEFD) showed significant negative correlations with 100-seed weight, 100-pod weight, YPP, average weight per pod, and shelling percentage. Principal component analysis (PCA) suggested that flowering redundancy traits as well as yield-related traits contributed more to the first three PCs when compared to other traits. This study addresses the lack of peanut flowering phenotypic and genetic diversity and lays the foundation for in-depth research on redundancy-related genes in peanut flowering.

Background: Sclerotium rolfsii, the causal agent of peanut southern blight, has become increasingly prevalent and harmful in China, causing serious economic losses to the peanut industry. To effectively manage peanut southern blight, this study evaluated the bioactivity of the new-generation sterol demethylation inhibitor (DMI) fungicide mefentrifluconazole against peanut S. rolfsii.Results: In this study, the DMI fungicide mefentrifluconazole exhibited excellent inhibitory activity against the mycelial growth of S. rolfsii, with a mean EC 50 value of 0.21 ± 0.11 mg L −1 and a range of 0.02 to 0.55 mg L −1 for 261 isolates collected from Hebei, Henan and Shandong provinces. Mefentrifluconazole significantly reduced the biomass of mycelia and affected the morphology of hyphae. Although sclerotia were more tolerant to mefentrifluconazole than mycelial growth, mefentrifluconazole greatly inhibited the formation and germination of sclerotia. In addition, sclerotia produced by mefentrifluconazole-treated mycelia were deficient in nutrients (e.g., protein, carbohydrate and lipid). These results indicated that mefentrifluconazole may reduce the population of S. rolfsii in the following year. In greenhouse experiments, mefentrifluconazole showed control efficacy and good persistence against peanut S. rolfsii. The preventative and curative activities of mefentrifluconazole at 200 mg L −1 against southern blight still reached 95.36% and 60.94%, respectively, after 9 days of application. No correlation was observed for the sensitivity of S. rolfsii to mefentrifluconazole and the tested DMI, quinone outside inhibitor and succinate dehydrogenase inhibitor fungicides. Conclusion: All data indicated that mefentrifluconazole could provide favorable control efficacy against S. rolfsii from peanuts and reduce the infection and population of S. rolfsii in the following year.

Abstract. Nurhalimah, Muhibuddin A, Syib’li MA. 2022. Screening of yeast isolates for the biocontrol of Sclerotium rolfsii. Biodiversitas 23: 3820-3826. Sclerotium rolfsii is a fungal pathogen that can reduce peanut production. Biological control with yeast antagonist offers an environment friendly control for this pathogen. The aim of this study was to isolate and identify yeast antagonists, and to study their antagonistic mechanism and potential to control the peanut disease caused by S. rolfsii. The study was conducted from October 2020 to May 2021. The study consisted of four stages: isolation and identification of yeasts, yeasts antagonist testing against S. rolfsii, molecular identification of yeast NH4, and scanning electron microscope observation of NH4 against S. rolfsii. Based on the research, four different yeast isolates were isolated, identified, and found able to be developed as a biological control agent to inhibit the growth of S. rolfsii. Identification showed that NH1 was Saccharomyces sp. with inhibitory percentage of 9,26%, NH2 was Candida sp. with 22,21% inhibition of, NH3 Pichia sp. with 11,11% inhibition of, and NH4 was Wickerhamomyces sp. with an inhibitory percentage of 32,96%. The results of the yeast antagonism mechanism showed that NH1 (Saccharomyces sp.) and NH3 (Pichia sp) had not showed any mechanism while, NH2 (Candida sp.) had a competition and NH4 (Wickerhamomyces sp.) had an antibiosis mechanism. NH4 can suppress the growth of S. rolfsii by more than 30%, which has been identified as species of Wickerhamomyces sp. (NH4) with 99.84% homology with Wickerhamomyces anomalus CBS 5759 with accession number MH545921.1.
