Characterization of a Factor from Rat Serum with Blocking Activity in the Passive Cutaneous Anaphylactic Reaction

Abstract: A blocking factor (BF) was obtained from normal and rat reaginic serum which inhibited the passive cutaneous anaphylactic (PCA) reaction of mouse and rat IgE in rat skin. Several chromatographic procedures on DEAE- and CM-cellulose at various pH values were compared in an attempt to obtain BF with high activity. BF-containing fractions after DEAE chromatography at pH 5,4 revealed two components on SDS-polyacrylamide gel: one at 50,000–60,000 and the other at 65,000–70,000 daltons. The latter component was also… Show more

“…In agreement with Theobald and König [14] our experiments showed that, whatever the procedure of purification used, the yield of the biological activity was low. The inhibitory activity was found only when the constant presence of a band of 76,000 daltons was always associated either to 2 bands (55,000 and 26,000 daltons in fraction C of P.50) or to a band corre sponding to the serum albumin (fractions SF5, SF6, SF7 of P.70).…”

“…About the purification of the BF, Theobald and König [14] showed that their BF found in normal rat and reaginic serum was characterized as a 70,000-dalton molecule that migrates or comigrates with the al bumin fraction. They have clearly shown that inhibi tion was neither due to albumin nor IgG-related.…”

“…It has been established in the past that sera of var ious mammalian species can inhibit the P CA reaction and the histamine release from rat mast cells and hu man basophil leukocytes in vitro and in vivo [1][2][3][4], Recently, our studies were directed towards the isola tion o f this molecule from human serum. It appears that at least two factors can mediate this reaction.…”

“…During the last decade, several studies were re ported with regard to serum factors with modulatory activity for histamine release in vivo and in vitro [1][2][3][4], For rat serum the active molecule was identified as a protein o f approximately 70,000 daltons and an IEP at pH 4.8 [2]. In human serum using the same pu rification procedure a protein with very similar char acteristics was obtained.…”
