Characterization of a dITPase from the hyperthermophilic archaeon Thermococcus onnurineus NA1 and its application in PCR amplification

Abstract: In this study, we found that deoxyinosine triphosphate (dITP) could inhibit polymerase chain reaction (PCR) amplification of various family B-type DNA polymerases, and 0.93% dITP was spontaneously generated from deoxyadenosine triphosphate during PCR amplification. Thus, it was hypothesized that the generated dITP might have negative effect on PCR amplification of family B-type DNA polymerases. To overcome the inhibitory effect of dITP during PCR amplification, a dITP pyrophosphatase (dITPase) from Thermococcu… Show more

“…Literature reports of increased PCR yield when a thermostable inorganic pyrophosphatase (IPPase) was included in the reactions inspired us to test this enzyme in a qPCR series to see if we could drive the reaction forward by degrading the pyrophosphate, one of the two products of the chain reaction [17], [18]. Unexpectedly, the addition of IPPase reduced the apparent reaction yield ( Figure 5A ).…”

“…T. onnurineus NA1 is a recently identified hyperthermophilic archaeon that was isolated from geothermal sediments from a black smoker on the ocean floor. 16 Several enzymes such as PPase17 (80°C), dITPase18 (75°C), histidinol phosphate phosphatase19 (80°C), and fructose‐1,6‐bisphosphatase20 (95°C) from T. onnurineus have been reported to have unique thermophilic enzymatic activities. A major interest in thermophilic enzymes concerns the development of specific physicochemical regulatory functions under harsh conditions while maintaining enzyme stability.…”
