Characterization of a Cellobiose Phosphorylase from a Hyperthermophilic Eubacterium,Thermotoga maritimaMSB8

Abstract: The cepA putative gene encoding a cellobiose phosphorylase of Thermotoga maritima MSB8 was cloned, expressed in Escherichia coli BL21-codonplus-RIL and characterized in detail. The maximal enzyme activity was observed at pH 6.2 and 80 degrees C. The energy of activation was 74 kJ/mol. The enzyme was stable for 30 min at 70 degrees C in the pH range of 6-8. The enzyme phosphorolyzed cellobiose in an random-ordered bi bi mechanism with the random binding of cellobiose and phosphate followed by the ordered releas… Show more

“…CbP and CdP have been purified from C. thermocellum (4)(5)(6)(7)40), and the presence of one or both of these enzymes has also been documented in other cellulolytic bacteria, including Ruminococcus flavefaciens (9), C. gilvus (36), R. albus (27), Prevotella ruminicola (26), C. stercorarium (34) and Thermotoga neapolitana (45), and T. maritime (33). Although C. thermocellum grows at ϳ60°C, specific activities of CbP and CdP have been measured at 37°C in prior studies (6)(7)(8)43).…”

“…The kinetic parameters of the C. uda CP E649C variant on methyl β-glucoside are better than those of Thermotoga maritima CP, which is the only known CP active on methyl β-glucoside [25]. For this enzyme a K m value of 135 mM and a k cat value of 6.5 s…”

“…2). These results indicate that the phosphorolytic reaction of GalGlyNAcP Vv followed the sequential bi-bi mechanism, like the reactions of inverting phosphorylases, such as GalGlyNAcP from B. bifidum (5), GalGlyNAcP Bl (27), GalGlyNAcP Cp (27), maltose phosphorylase (39), cellobiose phosphorylase (15,17,28,34), chitobiose phosphorylase (12), and laminaribiose phosphorylase (18).…”
