Characterization, anticancer drug susceptibility and atRA-induced growth inhibition of a novel cell line (HUMEMS) established from pleural effusion of alveolar rhabdomyosarcoma of breast tissue

Abstract: We recently established a cell line derived from pleural effusion from a 13-year-old girl with primary alveolar rhabdomyosarcoma (RMS with a chromosomal translocation t[2;13]) in the breast tissue. The cell line was designated as HUMEMS. Cases of primary alveolar RMS swelling in the breast are extremely rare (about 0.2% of all RMSs). Therefore, the HUMEMS cell line is an important material for studying therapeutics for malignant tumors in children. The HUMEMS cell line we isolated consisted of two morphologica… Show more

“…They are described below and summarized in Table 1. There are also 16 human RMS cell lines that have been described and used by single research groups (5–17); these are listed in Table 2. [Of note, during revisions of this article an independent list of human and murine RMS cell lines was published (18). ]…”

“…In the current study, we found that the NUTOS cells had the translocation t(2;13) (p11;q14). Ohi has also reported finding translocation t(13;19)(q14;p13) in the alveolar type of breast rhabdomyosarcoma cells (HUMENS) 11 …”

“…These large multinucleated cells resemble skeletal muscle cells, as they contract automatically. While rhabdomyosarcoma cells (HUMENS) that are derived from the alveolar type of breast tissue have myotubes and myofilaments, the NUTOS cells only have myofilaments 11 …”

“…So far, cell lines have been obtained from sarcomas mainly by two methods; seeding material from pleural effusions [5][6][7][8][9][10][11][12] or seeding of cells after enzymatic digestion of tumor material. [13][14][15][16][17][18][19] As we assumed it to be difficult to optimize enzymatic digestion protocols for each single tissue type affected by sarcoma and because pleural effusion is mainly performed in patients with end-stage lung metastases only, we explored here the potential to obtain patient-derived cell lines from sarcoma patients using the explant technique.…”
