Characterization and tissue-specific expression of the rat basic fibroblast growth factor antisense mRNA and protein

Abstract: An RNA transcribed from the antisense strand of the FGF-2 gene has been implicated in the regulation of FGF-2 mRNA stability in amphibian oocytes. We have now cloned and characterized a novel 1.1-kb mRNA (fgf-as) from neonatal rat liver.

“…An antisense transcript regulates its expression (Nudt6), functioning as a repressor (Knee et al, 1997; MacFarlane et al, 2010; MacFarlane and Murphy, 2010). There are also various transcription factors that can bind its promoter elements, such as HoxA10, AP-1 and SP-1 (Shah et al, 2012; Shibata et al, 1991).…”

“…Here, a decrease in FGFR1 expression was also accompanied by a decrease in glial proliferation in the prefrontal cortex, and the latter effect could be prevented by treatment with FGF2. In keeping with the idea of a mechanism for these effects (other than downstream signaling), FGF2 has an antisense transcript (FGF2-AS) that is known to regulate the expression of FGF2 (Knee et al, 1997; MacFarlane et al, 2010; MacFarlane and Murphy, 2010). Furthermore, corticosterone administration can differentially regulate FGF2 and FGF2-AS expression in both escapable and inescapable shock paradigms (Frank et al, 2007).…”

“…The following are known mammal cDNA sequences reported to accompany antisense transcripts selected from the recent literature and used in this study: Fgf2 (Knee et al 1997), Hif1a (Thrash-Bingham and Tartof 1999), Tgfb2 (Coker et al 1998), Tnfrsf17 (Hatzoglou et al 2002), Mkrn2 (Gray et al 2001), Hfe (Thenie et al 2001), Klhl1 (Benzow and Koob 2002), Ucn (Shi et al 2000), Wt1 (Moorwood et al 1998), Tnni1 (Podlowski et al 2002), Thra (Hastings et al 2000), Hsp70.2 (Murashov and Wolgemuth 1996), Hoxd-3 (Bedford et al 1995), Tyms (Dolnick 1993), CD3z (Lerner et al 1993), N-myc (Krystal et al 1990), Trp53 (Khochbin and Lawrence 1989), Ercc1 (van Duin et al 1989), and Hoxa11 (Hsieh- Li et al 1995). To identify the known sense-antisense transcripts in the bidirectional transcription pairs calculated in this article, we looked up the GenBank# or Refseq# of the known transcripts in the redundant version of our pair list.…”

“…Nudt6 is synthesized from the complementary strand of Fgf2, and was implicated to function in post-transcriptional regulation of Fgf2 [24]. Studies on Nudt6 are very limited, and its roles are still unknown [25]. This study demonstrates the expression of Nudt6 during brain development for the first time, and its relatively high expression implies that it might have various crucial roles during CNS development (Table  2).…”
