Characterization and genomic study of “phiKMV-Like” phage PAXYB1 infecting Pseudomonas aeruginosa

Abstract: Bacteriophage PAXYB1 was recently isolated from wastewater samples. This phage was chosen based on its lytic properties against clinical isolates of Pseudomonas aeruginosa (P. aeruginosa). In the present study, characterized PAXYB1, clarified its morphological and lytic properties, and analyzed its complete genome sequence. Based on the morphology of PAXYB1, it is a Podoviridae. The linear GC-rich (62.29%) double-stranded DNA genome of PAXYB1 is 43,337 bp including direct terminal repeats (DTRs) of 468 bp. It … Show more

“…The adsorption constant of phage Pf-10 calculated using the equation described by Kropinski [45] is (3.59 ± 1.42) × 10 −9 mL/min. The adsorption rate of phage Pf-10 was similar to that of other reported Pseudomonas-infecting phages of the Autographiviridae family, namely: Pseudomonas phage Andromeda (1.48 × 10 −9 mL/min) [63] and Pseudomonas phage PAXYB1 (3.70 × 10 −9 mL/min) [64].…”

“…PhiKMV-like viruses are lytic phages that infect Pseudomonas aeruginosa. They are members of the family Podoviridae with short noncontractile tails and double stranded DNA (Yu et al 2017). Another important Podoviridae viruses are T7like viruses.…”

“…Five test tubes containing 10 9 phage particles were immersed in different temperature water baths for approximately 1 h (4, 25, 37, 45, and 50 • C). Thereafter, the phage titers of all the samples were determined using the double-layer agar method (Yu et al, 2017).Three independent repeated experiments were performed. The average value was used to generate the figure, and the standard error of the mean (SEM) was marked.…”
