Characterization and discrimination of nasopharyngeal carcinoma and nasopharyngeal normal cell lines using confocal Raman microspectroscopy

Abstract: Abstract. Raman microspectroscopy can provide molecular-level information about the biochemical composition and structure of cells and tissues with excellent spatial resolution. In this study, Raman spectroscopy of individual cells from nasopharyngeal carcinoma cell lines C666-1, CNE2 and nasopharyngeal normal cell line NP69 are investigated for their differences. The spectral intensity ratio at 1449 and 1657 cm −1 with a decision line of I 1449 /I 1657 = 1.10 can very easily separate the tumor and normal cell… Show more

“…In comparison with the findings of Ye et al (2011), these 4 Raman spectra also exhibited a similar spectral profile indicating they are composed of the same components with subtle differences and variation occurring in certain bands as discussed in detail below. Band positions and their corresponding assignments are shown (Table 1).…”

“…For example, compared with normal cell NP69, the 3 NPC cell lines' Raman spectra exhibited some similar characterizations in comparison with Ye et al (2011) as follows. (i) The intensity of the band at 1003 cm −1 assigned to phenylalanine is stronger in NPC cell lines including CNE1, indicating variations in protein contribution to the different cell lines.…”

“…The obvious bands of CNE1 and CNE2 have been reported: [9][10] (2800-3030 cm -1 ). This was consistent with the theory that CNE1…”

“…Recently, the discrimination of NPC including cell lines and tissue [9][10][11][12] by RS was complemented. Spectral differences were observed between the cancer and normal subjects; Nine Raman band ratios were used for classification [10] using principal component analysis (PCA) and linear discriminant analysis (LDA), the sensitivity and specificity were reported to be 92% and 82%, respectively.…”
