Characterization and Application of Lytic Bacteriophages against Campylobacter jejuni Isolated from Poultry in Japan

Abstract: The aim was to isolate Campylobacter jejuni-specific lytic phages from meats on the market in Japan. These phages were effectively isolated from 13 of 15 86.7% retail chicken meat samples skin and liver by the enrichment method using Preston Campylobacter Selective Enrichment Broth and 10 host Campylobacter strains. Among the 26 phage isolates, 14 were extracted by means of C. jejuni L26 as a host strain. Phage PHC10 showed the broadest lytic spectrum: active against 67.4% of the 46 C. jejuni strains tested. “…What needs to be addressed in research studies is using local isolates of C. jejuni for selection of phages to increase their diversity. For example, for enrichment of phage isolates, Furuta et al, who found that phage susceptibility profiles of the Japanese Campylobacter strains were different from those of non-Japanese ones, suggest using locally isolated C. jejuni strains [70]. Previous research of Campylobacter phages relied heavily on just a few C. jejuni strains, notably NCTC 12662, which was frequently used for propagation and enrichment of phages, as discussed previously in this work.…”

“…For this purpose, 5 ml of each phage suspension in TM buffer was adsorbed onto glow‐discharged carbon‐coated 200 mesh copper grids, and then, after 3 min adsorption, the excess lysate was collected and the slides were negatively stained with 2% uranyl acetate (pH 4.0) for 15 s. The slides were examined using a Philips CM 100 transmission electron microscope at 12,000–80,000× magnification, and selected phages were recorded using iTEM software (Olympus Soft Imaging Solution). The bacteriophages were classified according to Carvalho, Gannon, et al (), Carvalho, Susano, et al () and Furuta et al ().…”

“…Sampling: Campylobacter phages are normally found in all places at which their hosts occur [50]. They have been isolated from poultry (i.e., meat, liver, skin, feces, and intestines), manure, and sewage [22,36,43,50,51,52]. High rates of Campylobacter phages have been isolated from organic farms [53].…”
