Antimicrobials are excessively used in Pakistan during broiler poultry production, however, status of antimicrobial resistance (AMR) is not known. We report on the frequency of extended spectrum β-lactamase (ESBL)-producing-Escherichia coli recovered from samples obtained from poultry farm environment (n=100) and chicken meat samples (n=100) collected at sales point of live bird market in District Peshawar, Pakistan. Phenotypically confirmed ESBL producers were further investigated for the presence of ESBL-and carbapenemase-encoding genes and types of integrons. Our results showed that 38.8% (14/36) and 47.6% (20/42) of E. coli recovered from farm environment and chicken meat, respectively, were found to be ESBL producers. PCR based results showed that 30% (6/20), 10% (2/20) and 20% (4/20) of these E. coli from chicken-meat were harboring blaCTXM, blaCTXM-1 and blaSHV2, respectively, while 20% were carrying blaOXA-48 and blaNDM-1. On the other hands, 28.5% (4/14), 28.5% (4/14) and 14.2% (2/14) of E. coli isolated from chicken farm-environment were harboring blaCTXM, blaCTXM-1 and blaSHV2, respectively, while, 28.5% (4/14) and 14.2% (2/14) were carrying blaOXA-48 and blaNDM-1. Integron 2 was PCR amplified from 17 isolates and integron 1 from 16 isolates, while, integron 3 was absent from all. Finally, insertion sequence ISCR1 was PCR-amplified from 15 isolates (41.6%), which was found associated with blaCTXM among 6 (16.6%) isolates suggesting its role in mobilization. Overall, our results suggest a high rate of occurrence of ESBL-and carbapenemase-carrying E. coli in poultry farm environment and chicken meat.

Literature regarding multidrug resistant (MDR) carbapenemase-producing Escherichia coli (CPE) recovered from food-producing animals is lacking from Pakistan. Here, we report on the isolation and characterization of MDR-CPE recovered from retail-poultry meat in Khyber Pakhtunkhwa Pakistan. A total of 101 retail poultry-meat samples were screened for Escherichia coli and tested against 17 different antibiotics through disc diffusion test to define MDRs. MDR isolates were genotyped by PCR for carbapenemase-encoding elements. Finally, integron typing, insertion sequence common region 1 (ISCR1) and its association with carbapenemaseencoding genes was established through PCR. Of the 33 E. coli isolates, 28 (Phylogroup B2=39.2% (11/28), A=32.1 % (9/28) and group D=28.5% (8/28) were MDR displaying resistance to cefotaxime, tetracycline, ampicillin and trimethoprimsulfamethoxazole, while, 96.4, 92.8, 89.2 and 85.7% were also found resistant to colistin-sulphate, enrofloxacin, doxycycline and ciprofloxacin, respectively. The blaVIM was predominantly identified in 8 (28.5%) isolates followed by blaNDM 4/28 (14.2%) and 2 isolates were carrying a combination of blaVIM+blaNDM. Class-1 integron was detected in 21 (75%), class-2 and class-3 integrons were detected in 25% and 7.1%, respectively. ISCR1 element was identified among 11 isolates (39.28%) that was found associated with blaVIM gene among 3 isolates (27.25%) suggesting involvement in its mobilization. We report the incidence of E. coli carrying carbapenemase-encoding genes located on the plasmids carrying integron 1 recovered from retail-poultry-meat in Mardan, Pakistan.

The prevalence of antimicrobial resistance (AMR) is increasing in human and animal pathogens, becoming a concern worldwide. However, prevalence and characteristics of AMR of bovine mastitis pathogens in large Chinese dairy herds are still unclear. Therefore, our objective was to determine the AMR profile of bacteria isolated from clinical mastitis in large (>500 cows) Chinese dairy herds. A total of 541 isolates of the 5 most common species, Staphylococcus aureus (n = 103), non-aureus staphylococci (NAS; n = 107), Streptococcus species (n = 101), Klebsiella species (n = 130), and Escherichia coli (n = 100), isolated from bovine clinical mastitis on 45 dairy farms located in 10 provinces of China were included. Presence of AMR was determined by minimum inhibitory concentrations using the microdilution method. Prevalence of multidrug resistance (resistance to >2 antimicrobials) was 27% (148/541). A very wide distribution of minimum inhibitory concentrations was screened in all isolates, including Staph. aureus isolates, which were resistant to penicillin (66%). In addition, NAS (30%) were more resistant than Staph. aureus to oxacillin (84%), penicillin (62%), tetracycline (34%), and clindamycin (33%). Prevalence of resistance to tetracycline was high (59%) in Streptococcus spp. Additionally, prevalence of resistance of both E. coli and Klebsiella spp. was high to amoxicillin/clavulanate potassium (81 and 38%, respectively), followed by tetracycline (only Klebsiella spp. 32%). A high proportion (27%) of isolates were multidrug resistant; the most frequent combinations were clindamycin-cefalexin-tetracycline or enrofloxacin-cefalexin-penicillin patterns for Staph. aureus; enrofloxacin-oxacillin-penicillin-tetracycline patterns for NAS; clindamycin-enrofloxacin-tetracycline patterns for Streptococcus spp.; amoxicillin/clavulanate potassiumceftiofur-polymyxin B patterns for Klebsiella spp.; and amoxicillin/clavulanate potassium-ceftiofur-polymyxin B patterns for E. coli. Resistance for 4 kinds of antimicrobials highly critical for human medicine, including daptomycin, vancomycin, imipenem, and polymyxin B, ranged from 0 to 24%. In conclusion, prevalence of AMR in mastitis pathogens was high on large Chinese dairy farms, potentially jeopardizing both antimicrobial efficacy and public health. Results of this study highlighted the need for improvements in antimicrobial stewardship and infection control programs in large Chinese dairy farms to reduce emergence of AMR.
