Characterisation of three fungal glucuronoyl esterases on glucuronic acid ester model compounds

Abstract: The glucuronoyl esterases (GEs) that have been identified so far belong to family 15 of the carbohydrate esterases in the CAZy classification system and are presumed to target ester bonds between lignin alcohols and (4-O-methyl-)d-glucuronic acid residues of xylan. Few GEs have been cloned, expressed and characterised to date. Characterisation has been done on a variety of synthetic substrates; however, the number of commercially available substrates is very limited. We identified novel putative GEs from a wid… Show more

“…Feruloyl esterases are also applied for improving the quality of bread, preparation and clarification of juices, synthesis of oligosaccharides and production of medicinal compounds (Fazary and Ju 2013). Glucuronoyl esterases are active against a wide range of natural and synthetic lignin-carbohydrate complexes (Hüttner et al 2017). Specifically, glucuronoyl esterases cleave ester linkages between the lignin and glucuronoxylan complexes (Arnling Bååth et al 2016).…”

“…xylanases-to reach their targets within the plant biomass. To date, all characterized GEs are either singledomain proteins, or are coupled with an N-terminal carbohydrate-binding module 1 (CBM1) domain [36]. This module is thought to allow the enzyme to bind easier to insoluble substrates, and thus increase the overall activity [37].…”

“…Glucuronoyl esterase (GE) is a recently defined carbohydrate esterase that has been isolated from several microorganisms [1][2][3][4][5][6]. GE has potential for production of biofuels and biomaterials in lignocellulose biorefineries [7][8][9][10][11][12]. GE cleaves alkali-labile bonds from the lignincarbohydrate complexes at acidic pH [12].…”

GE cleaves alkali-labile bonds from the lignincarbohydrate complexes at acidic pH [12]. Recent reports have proven that the removal of glucuronic acid branches from the hemicellulose significantly improved release of fermentable sugars for biofuels production [7,8,13].…”
