Characterisation of selected drugs with nitrogen-containing saturated ring structures by use of electrospray ionisation with ion-trap mass spectrometry

Abstract: The electrospray ionisation-ion-trap mass spectrometry (ESI-MS(n)) of selected drugs with nitrogen-containing saturated ring structures has been investigated. Sequential product-ion fragmentation experiments (MS(n)) have been performed to elucidate degradation pathways for the [M+H](+) ions and their predominant fragment ions. These MS(n) experiments result in characteristic fragmentations in which functional groups are generally cleaved from the ring systems as neutral molecules such as H(2)O, amines, alkenes… Show more

“…1 ion of cocaine at m/z 304.1 underwent insource fragmentation to give a signal at m/z 182.1 (relative abundance 40%), a signal that is also observed at MS 2 [44]. This corresponded to loss of the benzoic acid substituent as a neutral molecule, leaving a secondary carbonium ion.…”

“…With respect to recent CE-ESI-MS publications concerning some of the selected drugs as studied by Smyth et al [44], CE-ESI-MS 2 has been shown to be capable of identifying comigrating compounds, e.g., the comigration of amphetamine with nicotine in a study by Ramseier [27]. Zhu et al [45] have described a colloidal graphite-coated emitter for sheathless CE/nanoESI-MS. Baseline separation and structure elucidation of risperidone and 9-hydroxyrisperidone was achieved by coupling pressureassisted CE to ESI-TOFMS using the described sheathless electrospray emitter with a bare fused-silica capillary at pH 6.7.…”

“…Smyth et al [19] have carried out ESI-MS n studies on selected drugs with nitrogen-containing saturated ring structures including nicotine, olanzapine, risperidone, narcotine, reserpine, yohimbine, pethidine, prazosin and sidenafil. Supported by QTOF-MS/MS data, they have shown certain characteristic fragmentations in that functional groups were generally cleaved from the ring systems as neutral molecules such as H 2 O, amines, alkenes, esters, carboxylic acids etc.…”
