Every year, ten thousand Sunda Pangolins (Manis javanica) are rescued and confiscated from the illegal trade around Southeast Asia, including Thailand. Most of them are usually moribund and need intensive veterinary interventions to recover their health before being released to the wild. However, a lack of reliable haematology and clinical biochemistry references for Thai confiscated Sunda pangolins is a major difficulty for reaching this goal. The aim of this study was to establish a standard panel of haematology and serum biochemistry profiles used for confiscated pangolins in Thailand. Coccygeal venepuncture of 52 confiscated pangolins was performed. Haematology and serum biochemistry data acquisitions were made using automated blood analysers. The lower and upper limits at 90% confidence interval were calculated. The means of all parameters were calculated and then compared with those in two previous studies (p < 0.005). Blood corpuscle morphometry was done manually. All blood corpuscles in females were bigger than males. There were differences of haematologic and biochemical parameters among this study and previous studies in some aspects. Wide scientific discussion was made to explain these variations. A panel of haematology and serum biochemistry profiles for Thai confiscated Sunda pangolin has been established. Veterinarians can refer to all parameters to evaluate the animals’ health and diseases.

We attempted to characterize the cytochemical staining patterns of leukocytes and to determine the percentages of alpha-naphthyl acetate esterase (ANAE) and acid phosphatase (ACPase) positive lymphocytes in peripheral blood of thoroughbred foals at different ages. Blood samples were obtained from the jugular veins of 60 healthy thoroughbred foals, 1 day, 3 days and 1 year old. Each age group included 10 male and 10 female animals. Peroxidase (PO) activity was detected in neutrophils, eosinophils and monocytes. Lymphocytes were negative for PO staining. Periodic acid-Schiff (PAS) staining was observed in neutrophils and monocytes; eosinophils were negative. The majority of lymphocytes were negative, but a few cells showed granular PAS positivity. Monocytes were strongly positive for ANAE and ACPase, and the enzymatic reaction was common in lymphocytes. Neutrophils showed a weakly positive reaction for ANAE and ACPase. Eosinophil granules were negative or weakly positive for ANAE and usually negative for ACP, but intergranular areas were positive. Mean ANAE positive peripheral blood lymphocytes (PBL) were 67.70, 73.10 and 49.20% in females; 64.00, 70.53, and 50.60% in males 1 day, 3 days, and 1 year old, respectively. ACPase positive PBL were 27.33, 32.83, and 37.40% in females; 29.67, 31.67, and 38.40% in males 1 day, 3 days, and 1 year old, respectively. For both enzymes, the differences between mean values for the genders were not statistically significant, but significant differences were found with regard to age. We provide comparative hematological data as a guide for identifying blood cells in thoroughbred foals.

Blood analysis has recently become a popular tool to assess the welfare of the wild cats. However, the estimates of blood parameters may depend on the sampling method. We have tested (1) if the sampling procedure influences blood parameters and (2) what parameters are the most efficient in assessing the physiological status in wild cat species. We assessed the effect of handling time on red blood cells (RBC) and white blood cells (WBC) counts, the ratio of neutrophils to lymphocytes (N/L ratio), and serum cortisol level within 1 hr after the capture of the animal in six far-east wild cats (Prionailurus bengalensis euptilura). Also, we analyzed literature data in 17 cat species to assess the effect of place of study, type of immobilization, and handling time on WBC count and N/L ratio. Serum cortisol level varied significantly with the handling time. RBC and WBC counts were strongly affected by the handling time. N/L ratio was very robust and did not depend on the handling time. However, the analysis of literature data has shown that the prolonged handling time (over 1 hr) and the type of immobilization significantly influence the N/L ratio, whereas the WBC count does not depend on any of considered factors. We conclude that while most blood parameters of cats are affected by routine handling time, the N/L ratio does not vary if the samples are collected within 1 hr after the capture of the animal. All other tested parameters should be treated with caution.
