Changing Perspectives on the Role of DnaA-ATP in Orisome Function and Timing Regulation

Abstract: Bacteria, like all cells, must precisely duplicate their genomes before they divide. Regulation of this critical process focuses on forming a pre-replicative nucleoprotein complex, termed the orisome. Orisomes perform two essential mechanical tasks that configure the unique chromosomal replication origin, oriC to start a new round of chromosome replication: (1) unwinding origin DNA and (2) assisting with loading of the replicative DNA helicase on exposed single strands. In Es… Show more

“…The R-type DnaA boxes (consensus 5′-TTATMCACA-3′) are bound by DnaA complexed with ATP or ADP (ATP–DnaA or ADP–DnaA) and serve as anchors for subsequent DnaA-binding low-affinity boxes [ 20 , 21 ]. The low-affinity sites are bound by ATP–DnaA, which is important for the synchronization and/or regulation of chromosome replication [ 22 , 23 , 24 ]. The low-affinity ATP–DnaA boxes differ by 3–6 bases from the R-type DnaA box [ 19 , 25 ].…”

“…In the following, the most important aspects of the ATP-DnaA dependent initiation step of DNA replication is shortly summarized. For further details on this complex field, see expert reviews ( Leonard and Grimwade, 2010 ; Katayama et al., 2017 ; Hansen and Atlung, 2018 ; Leonard et al., 2019 ).…”
