Changes of Large Molecular Weight Hyaluronan and Versican in the Mouse Pubic Symphysis Through Pregnancy1

Abstract: During pregnancy, the mouse pubic symphysis undergoes expansion and remodeling resulting in formation of a flexible and elastic interpubic ligament allowing passage of a term fetus. In the current study, we sought to identify and characterize components of the extracellular matrix that likely play an important role in elongation and flexibility of the interpubic ligament during parturition. Mouse pubic symphyses and interpubic ligaments collected at time points during pregnancy and postpartum were utilized to … Show more

“…37 Briefly, after deparaffinization and dehydration, sections were enzymatically treated with 0.05% of trypsin (pH 7.8) for 30 min at 37°C to facilitate antibody binding. Endogenous peroxidase activity was blocked with 1% H 2 O 2 and 1% BSA in phosphate buffer for 30 min.…”

“…The expansion of the symphysis pubis is seen for the fetus to pass through the birth canal in a comfortable way. For this expansion, the synthesis of versican and HA is increased, and proteinases, like ADAMTS1, are decreased in the ECM (37). Cells obtained from amniotic fluid from pregnant women with fetuses who have neural tube defects do not make the storage of collagen.…”

“…Tissues were then digested in 100 mM ammonium acetate with 0.0005% phenol red (pH 7.0) containing 0.25 mg/ml proteinase-K (Roche, Indianapolis, IN) for 4 h at 60 C. Proteinase K was inactivated by boiling and undigested tissues were pelleted by centrifugation. An aliquot of 12.5 l supernatant equal to 0.125 mg dry weight of digested cervix was processed for FACE analysis as described elsewhere with minor modifications (32,33).…”
