Changes in the Expression of Mitochondrial Morphology-Related Genes during the Differentiation of Murine Embryonic Stem Cells

Abstract: During embryonic development, cells undergo changes in gene expression, signaling pathway activation/inactivation, metabolism, and intracellular organelle structures, which are mediated by mitochondria. Mitochondria continuously switch their morphology between elongated tubular and fragmented globular via mitochondrial fusion and fission. Mitochondrial fusion is mediated by proteins encoded by Mfn1, Mfn2, and Opa1, whereas mitochondrial fission is mediated by proteins encoded by Fis1 and Dnm1L. Here, we invest… Show more

“…It has been reported that there is an affirmative correlation between these proteins to their gene expressions (Lee et al . 2020). In the present study, upregulation of Mfn‐2/Drp‐1 was observed in most of the groups with LPS‐induction compared with the control.…”

“…Mitochondria are highly dynamic organelles, moving around the cell through the microtubule network, constantly changing their morphology by fission and fusion processes which are tightly controlled to balance possible metabolic or environmental alterations [ 22 , 25 ]. Furthermore, mitochondria are very communicative with the other cellular organelles which manifest their importance for signaling cascades and normal cell function.…”

“…In one mouse model, Fis1 levels could influence a pluripotent cell’s ability to differentiate through its influence on De novo fatty acid synthesis which required mitochondrial fission ( 148 ). However, minimal changes in Fis1 expression occur during mouse embryonic stem cell differentiation ( 149 ), suggesting further work needs to be done to elucidate the role of Fis1 during cellular differentiation.…”
