Changes in Skeletal Muscle and Body Weight on Sleeping Beauty Transposon-Mediated Transgenic Mice Overexpressing Pig mIGF-1

Abstract: Insulin-like growth factor (IGF-I) is an important growth factor in mammals, but the functions of the local muscle-specific isoform of insulin-like growth factor 1 (mIGF-1) to skeletal muscle development have rarely been reported. To determine the effect of pig mIGF-1 on body development and muscle deposition in vivo and to investigate the molecular mechanisms, the transgenic mouse model was generated which can also provide experimental data for making transgenic pigs with pig endogenous IGF1 gene. We construc… Show more

“…Many studies have demonstrated that overexpression of IGF-1 in skeletal muscle can promote muscle fiber hypertrophy and muscle production. Gao used α-actin protein regulators to drive the expression of IGF-1 in mouse myoblasts and obtained transgenic mice with muscle hypertrophy [ 45 ]. Zou used the skeletal muscle-specific myosin light chain enhancer to upregulate the expression of IGF-1 specifically in skeletal muscle and obtained muscle hypertrophy mice [ 46 ].…”

“…However, with increasing age, the number of β-cells decreased, glucose tolerance was impaired, and fasting blood glucose levels increased. There are two types of INSR in zebrafish [ 103 , 104 ]. Gong et al and Yang et al knocked out the INSR gene in these two types of zebrafish, and the results showed that zebrafish with INSR gene knockout showed hyperglycemia and a compensatory increase in β cells.…”
