Changes in optical properties of electroporated cells as revealed by digital holographic microscopy

Abstract: Changes in optical and shape-related characteristics of B16F10 cells after electroporation were investigated using digital holographic microscopy (DHM). Bipolar rectangular pulses specific for electrochemotherapy were used. Electroporation was performed in an "off-axis" DHM set-up without using exogenous markers. Two types of cell parameters were monitored seconds and minutes after pulse train application: parameters addressing a of the cell (refractive index and cell height) and cell parameters (projected are… Show more

“…The ability to measure cell count was validated for the first time in 2008 by Mölder et al [21], using human breast and prostate cancer cell lines, as well as a mouse fibroblast cell line. Measurement of cell growth has been well documented for both adherent and non-adherent cells [8,9,15,[25][26][27][28]47,49,56,59,61]. By segmentation of cellular outlines, morphology and motility can also be studied [7,50,51,57,80].…”

“…Typically, some further restrictions are placed on the culture chambers used, as normal cell culture at these time scales requires regular change of the cell culture medium. Consequently, long term studies have used various kinds of perfusion chambers to be able to replace the medium without disturbing the imaging view [30,32,39,43,49,55,61]. Several studies have also used larger culture chambers with large enough volume to avoid changing the medium entirely, such as by using a T25 cell culture flask (Table 1).…”

“…For independent computation of these two parameters the decoupling procedure was applied, using two perfusion media with different refractive indices . Cell RI and height were computed on a square area of 3 × 3 pixels, identified by a matlab (Mathworks, Natick, MA, USA) code in the region of maximum phase shift, following the procedure described in our previous work .…”

“…A combination of phase‐shifting interferometry and optical interferometry with digital phase shifting technology has greatly improved the precision of quantitative phase measurement, but its application has been limited by the resulting interference signals . Digital holographic microscopy is a high accuracy measuring method related to optical interference . Nelson et al.…”
