Changes in electrophoretic mobilities of alpha-and beta-lipoproteins as a result of plasma delipidation.

Abstract: A two-phase system containing the ternary mixture butanol/disopropyl ether/plasma in different proportions yields ordered delipidation of alpha-lipoproteins, pre-beta-lipoproteins- and beta-lipoproteins in plasma, as quantitated by densitometry after electrophoresis. As a consequence of delipidation the electrophoretic mobilities of pre-beta-lipoproteins and beta-lipoprotein increased, that of alpha-lipoprotein decreased.

“…Plasma proteins, including apolipoproteins, and other chemical constituents, and enzyme activities, remained stable in the delipidated plasma. Minor changes in electrophoretic mobilities after in vitro delipidation of these plasma lipoproteins have also been reported by our group [2]. Furthermore, apolipoproteins in delipidated plasma were capable of dissolving extracellular cholesterol [3] and were also able to mediate cholesterol efflux in a wide variety of cells including smooth muscle cells and macrophages [4-lo].…”

“…LA induced higher levels of lipid-stainable material in pre-P-HDL levels in both hypercholesterolemic and normocholesterolemic animals. However, although densitometric scans of lipidstainable materials have been shown to correlate with lipids in lipoproteins after electrophoresis [2], not all the lipoproteins stain equally.…”

“…It has also been shown that PDP results in ordered delipidation. In plasma, HDL is first delipidated followed by VLDL and LDL [48]. All apolipoproteins in plasma are retained after PDP treatment, and the net effect of these apolipoproteins is RCT.…”
