Changes in Culture Expanded Human Amniotic Epithelial Cells: Implications for Potential Therapeutic Applications

Abstract: Human amniotic epithelial cells (hAEC) isolated from term placenta have stem cell-like properties, differentiate into tissue specific cells and reduce lung and liver inflammation and fibrosis following transplantation into disease models established in mice. These features together with their low immunogenicity and immunosuppressive properties make hAEC an attractive source of cells for potential therapeutic applications. However, generation of large cell numbers required for therapies through serial expansion… Show more

“…Moreover, fibroblasts have immunosuppressive properties comparable to MSCs [52]. Otherwise, those non-MSC cells may derive from a limited number of contaminating AECs which were not removed in the first enzymatic digestion [53]. These mesenchymal-type cells exhibited some properties typical for MSCs.…”

“…Membranes were then subjected to blunt dissection to separate the choriodecidua from the amnion. Amnion tissue was used to isolate amnion cells using methods adapted from three protocols (23)(24)(25). Briefly, amnion cells were digested with 0.25% trypsin-EDTA (Gibco) at 37 C for 30 min.…”

“…Interestingly, cells derived from either the epithelial or mesenchymal layers of human amniotic membrane-derived cells (HAMCs) impair lymphocyte activation, dendritic cell maturation, and inflammatory cytokine production in vitro (9)(10)(11)(12)(13). Moreover, treatment with total amniotic membrane, amniotic-derived cells, or their conditioned medium was found to be protective in different models of inflammatory fibrotic conditions (9,12,14,15).…”
