Changes in Circulating MicroRNAs Are Associated With Childhood Obesity

Abstract: This study provides the first evidence that circulating miRNAs are deregulated in prepubertal obese children. Thus, the very early detection of an abnormal circulating miRNA profile may be a promising strategy to identify obese children who may suffer from metabolic abnormalities.

“…Previous studies have shown that some differentially and abundantly expressed miRNAs (down-regulated miRNAs: ssc-let-7i, ssc-miR-21, ssc-miR-27b-3p and up-regulated miRNAs: ssc-miR-1, ssc-miR-133a-3p, ssc-miR-486) were responsible for regulating adipogenic differentiation [36][37][38][41][42][43]. ssc-miR-1 and ssc-miR133a were found extensively in skeletal muscle tissues while ssc-miR-21, ssc-miR-27b-3p, and the Let-7 family play important roles in adipomuscle and myocardial tissue.…”

“…Therefore, ssc-miR-1 and ssc-miR-133a may increase lipometabolism in pigs. In addition, the concentration of miR-486 in the serum is correlated with T2DM and obesity, and miR-486, as a circulating miRNA, may be related with metabolic disorders [43]. Thus, the up-regulation of ssc-miR-486 in Laiwu pigs may be a potential biomarker of obesity.…”

“…Soon thereafter, the first study on circulating miRNAs in the context of pediatric obesity was published by the same group [48]. Also performing Taqman array analysis, Prats-Puig et al [48] profiled the circulating miRNAs in prepubertal children.…”

“…Moreover, miRNAs have been reported to regulate the expression of VEGF (vascular endothelial growth factor) in the retina and to participate to the development of diabetic retinopathy [36]. Kovacs et al identified a set of up-regulated NF-κB-responsive miRNAs in the retina of diabetic rats, including miR-146a/b, miR-155, miR-132 and miR-21 [37]. MiR-155 is known to be involved in immuno-modulatory signaling, whereas miR-21 and mir146a/b participate to fibrotic responses and miR-132 to angiogenesis.…”
