Proline-rich tyrosine kinase 2 (Pyk2) is a member of the focal adhesion kinase (FAK) subfamily of cytoplasmic tyrosine kinases. The C-terminal Pyk2 focal adhesion-targeting (Pyk2-FAT) domain binds to paxillin, an adhesion molecule. Paxillin has five leucine-aspartate (LD) motifs (LD1–LD5). Here, we show that the second LD motif of paxillin, LD2, interacts with Pyk2-FAT, similar to the known Pyk2-FAT/LD4 interaction. Both LD motifs can target two ligand-binding sites on Pyk2-FAT. Interestingly, they also share similar binding affinity for Pyk2-FAT with preferential association to one site relative to the other. Nevertheless, the LD2–LD4 region of paxillin (paxillin133–290) binds to Pyk2-FAT as a 1:1 complex. However, our data suggests that the Pyk2-FAT and paxillin complex is dynamic and it appears to be a mixture of two distinct conformations of paxillin which almost equally compete for Pyk2-FAT binding. These studies provide insight into the underlying selectivity of paxillin for Pyk2 and FAK which may influence the differing behavior of these two closely-related kinases in focal adhesion sites.

Higher erythrocyte deformability may reduce the risk of circulatory diseases by enhancing oxygen delivery and reducing the load on the cardiovascular system. The effect of endurance training on erythrocyte deformability is not clear. This study explored the impact of endurance training on erythrocyte deformation and shape and investigated the underlying mechanisms of hemorheological alterations. Forty male Wistar rats were randomly divided into two groups: sedentary (S; n = 21) and exercised (E; n = 19). Hematological indices and erythrocyte shape were measured at the end of the 11th week. The gene expression of erythropoietin (Epo) and the Epo receptor (EpoR) was quantified using reverse transcription-PCR (RT-PCR), and Epo protein expression was analyzed using Western blotting. Endurance training significantly decreased the abnormality ratio of erythrocyte shape (P < 0.01). The deformability indicator (DI) of red blood cells was lower in the E group than in the S group (P < 0.01). Eleven weeks of endurance training increased Epo mRNA and protein expression in the kidney (P < 0.01), EpoR mRNA expression in the bone marrow (P < 0.05), and relative circulating Epo (P < 0.01) compared to the sedentary group. The decrease in the erythrocyte morphological index and the maximum of deformability indicator were associated with an increase in relative circulating Epo. In conclusion, 11 weeks of endurance training increased erythrocyte deformability. Epo and EpoR may contribute to the decreased morphological index and deformability indicator in erythrocytes during endurance training in rats.

Currently, it is unclear which index of haematological parameters could be used to most easily monitor iron deficiency during endurance training. To address this question, 16 male Wistar rats were randomly assigned to two groups: a sedentary group (n = 8) and an exercised group (n = 8). Initially, animals in the exercise group started running on a treadmill at a rate of 30 m/min, on a 0% grade, for 1 min/session. Running time was gradually increased by 2 min/day. The training plan was one session per day during the initial 2 weeks and two sessions per day during the third to ninth week. At the end of the 9-week experiment, we analysed the blood of the experimental animals for haemoglobin levels, erythrocyte numbers, haematocrit, serum iron levels, total iron binding capacity, transferrin saturation, serum ferritin levels and soluble transferrin receptor (sTfR) levels, and we calculated the ratio of sTfR/ferritin. Erythrocyte numbers, haemoglobin levels and haematocrit values were decreased after 9 weeks of exercise, but sTfR and sTfR/ferritin values were increased (P < 0.01 or P < 0.05). The training regime significantly increased TfR mRNA levels in the bone marrow cells of the exercised rats compared with the sedentary group (1.8 ± 0.5 vs. 1.1 ± 0.2, P < 0.01). These results revealed a significant correlation between TfR levels in the bone marrow cells and the ratio of sTfR/ferritin (r = 0.517; P < 0.01) and sTfR levels (r = 0.206; P < 0.05) in sedentary and exercised rats. In conclusion, we show that sTfR indices and the ratio of sTfR/ferritin could be useful indicators for monitoring iron deficiency during endurance training.
