Abstract. Ovarian cancer causes more deaths than any other malignant tumor of the female reproductive system. This is because the condition usually goes undetected until the late stages. The purpose of the present study is to identify alterations in the serum proteome profile during the development of ovarian cancer and to provide an experimental basis for discovering new and valuable serum biomarkers for the early detection of ovarian carcinoma. Surface-enhanced laser desorption/ionization-time of flight (SELDI-TOF-MS) was used to profile changes in the serum proteome of Fischer 344 rats with ovarian cancer during the progress of tumor development. Sera were collected from the rats on day A (1 week before injection of tumor cells), day B (4 weeks after injection), and day C (6 weeks after injection). Each sample was subjected to SELDI-TOF-MS testing. Peak detection and alignment and selection of peaks with the highest discriminatory power were performed using ProteinChip biomarker software. Decision tree analyses were performed using biomarker pattern software. Finally, 3 peaks were found to be the most valuable ones (3759, 4659 and 9318 Da). The expression frequency of m/z 3759-Da peaks was downregulated and another two frequencies (4659 and 9318 Da) were upregulated, and the levels of expression of these three proteins showed the same tendency as the expression frequency during the development of ovarian cancer. The total accuracy rate of diagnosis at 4 and 6 weeks post-injection was 94.7 and 97.3%, respectively. Profiling the serum proteome changes during the process of the cancer development using SELDI-TOF-MS may provide useful information regarding carcinogenesis and facilitate discovery of novel serum biomarkers for early detection.

Numbers of studies used surface enhanced laser desorption/ionization-time of flight mass spectrometry (SELDI-TOF-MS) to find novel serum biomarkers for non-small cell lung cancer (NSCLC). It is arguable whether the SELDI technique has its value of diagnostic accuracy for NSCLC. The purpose of our study is to determine the diagnostic accuracy of SELDI-TOF-MS for diagnosing NSCLC. The Cochrane Central Register of Controlled Trials, MEDLINE, Pub Med, EMBASE, the Chinese Biomedical Literature Database, the China Academic Journals Full-text Database, and the Chinese Scientific Journals Database were searched systematically for potential studies. Reference lists of included studies and review articles were also reviewed. All studies that reported data on patients with a confirmed diagnosis of NSCLC and that compared the measurement of SELDI-TOF-MS with pathology standard were considered for inclusion. 11 studies were included in the systematic review. The ranges of the diagnostic value of SELDI-TOF-MS for NSCLC were as follows: sensitivity (SEN) was 0.70~1.00; specificity (SPE) was 0.68~1.00; positive likelihood ratio (PLR) was 2.23~23.14; negative likelihood ratio (NLR) was 0.04~0.43; and diagnostic odds ratio (DOR) was 5.17~621.0, respectively. SELDI-TOF-MS showed high accuracy in identifying NSCLC, and could be a potential screening tool for diagnosing NSCLC patients.

Liver fibrosis is a serious disease that affects around 350-400 million people worldwide. The main approach for fibrosis staging is liver biopsy, which is an invasive procedure that is not endured pretty well by patients. Currently, some serum-based biomarker panels are available for diagnosis and staging of liver fibrosis. Recent high-throughput proteomic studies are also very promising for identification of novel biomarkers for diagnosis and/or treatment of liver fibrosis. We hereby review the application of proteomic profiling studies for identification of fibrosis biomarkers with their advantages and drawbacks.
