As a calcium-regulated protein, CaMK II is closely related to cell death, and it participates in the development of pathological processes such as reperfusion injury, myocardial infarction, and oligodendrocyte death. The function of CaMK II activation in acute pancreatitis (AP) remains unclear. In our study, we confirmed that the expression of p-CaMK II was increased significantly and consistently in injured pancreatic tissues after caerulein-induced AP. Then, we found that KN93, an inhibitor of CaMK II, could mitigate the histopathological manifestations in pancreatic tissues, reduce serum levels of enzymology, and decrease oxidative stress products. Accordingly, we elucidated the effect of KN93 in vitro and found that KN93 had a protective effect on the pancreatic acinar cell necroptosis pathway by inhibiting the production of ROS and decreasing the expression of RIP3 and p-MLKL. In addition, we identified the protective effect of KN93 on AP through another mouse model induced by pancreatic duct ligation (PDL). Together, these data demonstrated that CaMK II participates in the development of AP and that inhibiting CaMK II activation could protect against AP by reducing acinar cell necroptosis, which may provide a new idea target for the prevention and treatment of AP in the clinic.

Background: A root cause analysis (RCA) is a structured method used to address problems. It seeks to identify the root causes of problems, and proposes solutions to achieve an ultimate goal. This study sought to investigate the effects of an RCA in treating elderly patients with acute pancreatitis (AP).Methods: 94 patients with AP, who had been admitted to our hospital from January 2019 to January 2020, were enrolled in the study. The patients were divided into two groups using the random number table method. Each group comprised 47 patients. The control group received routine care, while the observation group underwent RCA care. The clinical efficacy, patients' negative emotions before and after the intervention, the incidence of complications, and patients' levels of satisfaction with the nursing they received were compared between the two groups.Results: The total clinical effective rate of the observation group was higher than that of the control group (P<0.05). The Hamilton Anxiety Scale and Hamilton Depression Scale scores of the observation group were lower than those of the control group after the 2-week intervention (P<0.05). The incidence of total complications in the observation group was lower than that in the control group (P<0.05). The observation group's level of satisfaction with the nursing they received was higher than that of the control group (P<0.05). Conclusions:The RCA method can improve clinical efficacy, relieve negative emotion, reduce the incidence of complications, and improve the level of satisfaction with nursing of elderly patients with AP. It is worthy of promotion.

Emodin (6-methyl-1,3,8-trihydroxyanthraquinone) is a naturally occurring anthraquinone derivative found in roots and leaves of various plants, fungi and lichens. For a long time it has been used in traditional Chinese medicine as an active ingredient in herbs. Among other sources, it is isolated from the rhubarb Rheum palmatum or tuber fleece-flower Polygonam multiflorum. Emodin has a wide range of biological activities, including diuretic, antibacterial, antiulcer, anti-inflammatory, anticancer and antinociceptive. According to the most recent studies, emodin acts as an antimalarial and antiallergic agent, and can also reverse resistance to chemotherapy. In the present work the potential therapeutic role of emodin in treatment of inflammatory diseases, cancers and microbial infections is analysed.
