cGMP signaling pathway that modulates NF-κB activation in innate immune responses

Abstract: The nuclear factor-kappa B (NF-kB) pathway is an evolutionarily conserved signaling pathway that plays a central role in immune responses and inflammation. Here, we show that Drosophila NF-kB signaling is activated via a pathway in parallel with the Toll receptor by receptor-type guanylate cyclase, Gyc76C. Gyc76C produces cyclic guanosine monophosphate (cGMP) and modulates NF-kB signaling through the downstream Tollreceptor components dMyd88, Pelle, Tube, and Dif/Dorsal (NF-kB). The cGMP signaling pathway comp… Show more

“…However, in the current study we couldn't observe the two lipogenic enzymes as nsP3 interacting partners in THP1 derived macrophages. In our study, cGMP-PKG signaling pathways were found to be modulated which directly regulate the NF Kappa beta pathway, facilitating the activation of innate immune responses (33). Surfeit 4, another interacting partner, is a well-known protein in HCV infection.…”

“…STAT3 is a transcription factor for fibrotic genes such as collagen I and α-SMA [ 42 , 54 ]. cGMP is generated as a result of relaxin activity, and its signaling pathway may inhibit NF- κB activation in innate immunity [ 95 ]. cGMP is cyclic guanosine monophosphate, ERK1/2 is extracellular-signal regulated kinase 1/2, NOS is nitric oxide synthase, RXFP1 is relaxin family peptide receptor 1, and TGFβ is transforming growth factor-β.…”

“…In D. melanogaster , the cGMP-dependent signaling pathway regulates NF-κB in the immune system. Engineered NPs exhibit a heightened ability to target DCs and promote the development of a tolerogenic phenotype during the induction of immune tolerance [ 97 , 98 ]. Table 5 summarizes the diverse mechanisms and pathways that offer promising targets for the therapeutic regulation and intervention of the immune system.…”
