Cerebrospinal Fluid MicroRNA Signatures as Diagnostic Biomarkers in Brain Tumors

Abstract: Central nervous system (CNS) malignancies include primary tumors that originate within the CNS as well as secondary tumors that develop as a result of metastatic spread. Circulating microRNAs (miRNAs) were found in almost all human body fluids including cerebrospinal fluid (CSF), and they seem to be highly stable and resistant to even extreme conditions. The overall aim of our study was to identify specific CSF miRNA patterns that could differentiate among brain tumors. These new biomarkers could potentially a… Show more

“…Biomarkers are important in oncology, and are more recently including miRNas (18,37). A few studies have pro led meningioma tumors, suggesting potential circulating biomarkers, including miRNAs, that could advance cancer diagnosis and prognosis (26,38,39). Genome-wide array screening revealed that miRNA clusters are implicated in meningioma tumorigenesis, correlating with proliferation and predicting recurrence of this disease (40).…”

“…miRNAs are initially transcribed as long primary miRNAs (pre-miRNAs), and are subsequently processed by the microprocessor complex that contains the double strand RNA-specific endoribonuclease DROSHA and its RNA binding partner DiGeorge Syndrome critical region gene 8 (DGCR8) (72). Mature miRNAs bind to multiple targets (73). miRNAs are frequently deregulated in many types of cancer and regulate tumor progression, metastasis, and anti-cancer drug resistance (74)(75)(76).…”

“…Several miRNA profiles have been proposed for GBM diagnosis by liquid biopsy isolation (i.e., as proposed by Kopkova and colleagues and by Ebrahimkhani et al), but no one has correlated their expression with PET imaging features [ 330 , 331 ].…”
