Cerebrospinal fluid amyloid β and tau in LRRK2 mutation carriers

Abstract: The disposition of Aβ and tau is likely important in both LRRK2-related and sporadic PD, even during early phases of the disease. A better understanding of their production, aggregation, and degradation, including changes in their CSF levels, may provide insights into the pathogenesis of PD and the potential utility of these proteins as biomarkers.

“…In this respect, most previous studies have focused on patients with more advanced disease and dementia. However, emerging evidence indicates that Aβ pathology may play a role in PD at earlier disease stages than previously thought 18 27. Recent post mortem studies report increased Aβ deposition in patients with non-tremor dominant disease onset in which PIGD and bradykinesia are the prominent motor symptoms 9 12…”

“…Assays were measured using the INNO-BIA AlzBio3 kit (Innogenetics, Gent, Belgium) as described previously [3]. We have extensively utilized the assay platform and quality control procedures in our laboratory [3,8,16,17]. We found that the concentrations of CSF Aβ 42 and tau in the DATATOP CSF samples, frozen for more than two decades, were comparable with CSF samples collected recently [8].…”

“…In particular, reduced striatal dopamine function strongly correlates with reduced CSF Aβ proteins3 rather than CSF α-synuclein levels4 in these preclinical LRRK2 cases. In sporadic PD, many studies have shown correlations between reduced CSF Aβ and cognitive impairment and more rapid cognitive decline,5–8 particularly with deficits in verbal fluency which may have a motor component.…”
