BackgroundThe brain biochemical changes of social anxiety have not been clarified although there have been a limited number of MR spectroscopic studies which utilized metabolite/creatine ratios. Present study aimed to explore the alteration of absolute metabolite concentration in social anxiety disorder using quantitative MR spectroscopy.Materials and MethodsWith a 3.0T MR scanner, single voxel MR spectroscopy (stimulated echo acquisition mode, TR/TE/TM = 2000/20/16 ms) was performed in the left dorsolateral prefrontal cortex and related regions of nine medication-free patients with social anxiety disorder and nine controls. Absolute metabolite concentration was calculated using tissue water as the internal reference and corrected for the partial volume of cerebrospinal fluid.ResultsIn the left dorsolateral prefrontal cortex, the N-acetyl aspartate/creatine ratio of patients was significantly higher than that of controls, and this was due to the decrease of creatine concentration instead of the increase of N-acetyl aspartate concentration. Furthermore, the creatine concentration of the left dorsolateral prefrontal cortex was negatively correlated with the scores of Liebowitz social anxiety scale.ConclusionsThe alteration of creatine level in the left dorsolateral prefrontal cortex suggests abnormal energy metabolism and correlates with symptom severity in social anxiety disorder. And metabolite concentration is preferable to metabolite/creatine ratio for the investigation of individual, absolute metabolite changes in this region of social anxiety disorder.

Although N-acetylaspartate (NAA) has long been recognized as the most abundant amino acid in neurons by far, its primary role has remained a mystery. Based on its unique tertiary structure, we explored the potential of NAA to modulate aggregation of amyloid-beta (Aβ) peptide 1-42 via multiple corroborating aggregation assays along with electron microscopy. Thioflavin-T fluorescence assay demonstrated that at physiological concentrations, NAA substantially inhibited the initiation of Aβ fibril formation. In addition, NAA added after 25 min of Aβ aggregation was shown to break up preformed fibrils. Electron microscopy analysis confirmed the absence of mature fibrils following NAA treatment. Furthermore, fluorescence correlation spectroscopy and dynamic light scattering measurements confirmed significant reductions in Aβ fibril hydrodynamic radius following treatment with NAA. These results suggest that physiological levels of NAA could play an important role in controlling Aβ aggregation in vivo where they are both found in the same neuronal compartments.

ZusammenfassungSeit Einführung der 7 Tesla (7 T) Ultrahochfeld-MRT (UHF-MRT) in die klinische Forschung werden ihre Anwendungsmöglichkeiten für die neuroradiologische Diagnostik evaluiert. Die 7 T MRT ermöglicht eine Verbesserung der strukturellen und funktionellen Bildgebung aufgrund ihres hohen Signalzu-Rausch-Verhältnisses, der erzielbaren räumlichen Auflösung und ihrer hohen Sensitivität für Suszeptibilitätsartefakte. Akquisitionszeit und Bildqualität erleiden jedoch Verluste durch lokale Magnetfeldinhomogenitäten und die Parameterwahl und Schichtanzahl werden aufgrund der höheren Energiedeposition im Gewebe negativ beeinflusst. Neben den technischen Grundlagen der 7 T UHF-MRT werden neuroradiologische Anwendungsmöglichkeiten bei Hirntumoren, entzündlichen und degenerativen ZNS-Erkrankungen, Epilepsie sowie zerebrovaskulären Erkrankungen aufgezeigt und Möglichkeiten der MR-Spektroskopie, funktionelle MRT und MR-Angiografie betrachtet. Bis zur Einführung der 7 T MRT in die klinische Diagnostik werden Optimierungen von Hard-und Software sowie klinische Studien notwendig sein, um ihre Vorteile für die neuroradiologische Diagnostik bestmöglich nutzen zu können.
