Ceratocystis pirilliformis, a New Species from Eucalyptus nitens in Australia

Abstract: Several species of Ceratocystis have been recorded on Eucalyptus. These include C. fimbriata, C. eucalypti, C. moniliformis and C. moniliformopsis. Of these, only C. fimbriata is known as a pathogen; it recently has been found causing serious wilt diseases in Uganda, Congo and Brazil. This study was undertaken to collect Ceratocystis species, including C. eucalypti, from artificially induced wounds on Eucalyptus nitens near Canberra in southeastern Australia. Trees were wounded in October 2000, and wounds were… Show more

“…Until the early 2000s, species of Ceratocystis s.s. were primarily defined based on unique morphological characteristics (Barnes et al 2003;Kile et al 1996;Upadhyay 1981;Wingfield et al 1996). In some cases, identification has been supplemented using mating studies which, for example, could be used to confirm the species boundaries between C. fimbriata, C. platani and C. cacaofunesta (Engelbrecht and Harrington 2005;Ferreira et al 2010).…”

“…Initial phylogenetic species delineation in Ceratocystis relied heavily on gene genealogies of the 5.8S and surrounding internal transcribed spacer regions 1 and 2 (ITS1 and ITS2) (Barnes et al 2003;Roux et al 2004). In an attempt to increase the resolution of species boundaries, sequences of the β-tubulin 1 (βT 1) and translation elongation factor 1-α (EF 1-α) gene regions have generally been used in combination with the ITS region for species delineation (Van Wyk et al 2012, 2011b.…”

“…Four isolates from Australia were obtained from artificially induced wounds made on the stems of A. mearnsii trees near Cann River in the state of Victoria, Australia, collected as part of a previous study (Barnes et al 2003). All cultures used in this study have been preserved in the culture collection (CMW) of the Forestry and Agricultural Biotechnology Institute (FABI), University of Pretoria, South Africa and representative cultures have also been deposited with the Centraalbureau voor Schimmelcultures, Utrecht, The Netherlands (CBS).…”
