Ceramide‐rich platforms in transmembrane signaling

Abstract: Recent evidence suggests that ceramide regulates stress signaling via reorganization of the plasma membrane. The focus of this review will be to discuss the mechanism by which acid sphingomyelinase (ASMase)-generated ceramide initiates transmembrane signaling in the plasma membrane exoplasmic leaflet. In particular, we review the unique biophysical properties of ceramide that render it proficient in formation of signaling domains termed ceramide-rich platforms (CRPs), and the role of CRPs in the pathophysiolog… Show more

“…Sphingolipids such as ceramide are currently recognized as vital regulators of many biological processes (1)(2)(3)(4), and the levels and turnover of these bioactive sphingolipids are regulated by several enzymes. The hydrolysis of sphingomyelin (SM) 2 is a major pathway for ceramide production, and this reaction is catalyzed by the sphingomyelinases (SMases).…”

“…The hydrolysis of sphingomyelin (SM) 2 is a major pathway for ceramide production, and this reaction is catalyzed by the sphingomyelinases (SMases). Currently, six different SMases have been identified and grouped based upon their catalytic pH optima, including an acid SMase, an alkaline SMase and four neutral SMases (N-SMases) (5)(6)(7)(8).…”

“…aSMase regulates anti-DENV NS1 Ab-induced ceramide generation, NO production, and apoptosis Ceramide can be generated from sphingomyelin hydrolysis by aSMase (41,43,46,47). Interestingly, after anti-DENV NS1 stimulation, the accumulation of aSMase could be detected by confocal microscopic observation (Fig.…”

“…The formation of lipid rafts, the membrane microdomains that are enriched in cholesterol and glycosphingolipids, play an important role in cellular signaling in response to various stimuli (31)(32)(33)(34)(35). Ceramide, which is enriched in lipid rafts, acts as a second messenger of multiple extracellular stimuli in cell survival, inflammation, and apoptotic cell death (36)(37)(38)(39)(40)(41). We investigated the formation of raft structure and the generation of cellular ceramide after binding of anti-DENV NS1 Abs to endothelial cells.…”

“…In previous studies, factors of inflammation, including TNF-α, ROS, LPS, amyloid-β, IL-1β, and LDL, stimulated SMase activation activity, which then produced ceramide and initiated ceramide signal pathways to transfer death signaling [25][26][27]. This study evaluated the effects of LMG on bacterial SMase activity in vitro.…”
