Central role of the TIR‐domain‐containing adaptor‐inducing interferon‐β (TRIF) adaptor protein in murine sterile liver injury

Abstract: Multiple pathways drive the sterile injury response in the liver; however, it is unclear how the type of cells injured or the mechanism of injury activates these pathways. Here, we use a model of selective hepatocyte death to investigate sterile liver injury. In this model, the TIR-domain-containing adaptor-inducing interferon-β (TRIF) was a central mediator of the resulting intrahepatic inflammatory response that was independent of both upstream Toll-like receptor (TLR) 4 signaling and downstream type I IFN s… Show more

“…A pre-amplification step was included before the multiplex qRT-PCR assay (Brempelis et al, 2017), which included PCR reactions of cDNA as templates, and a primer mixture of TaqMan assays of interest (Thermo Fisher Scientific, Waltham, MA, USA) and the BIO-X-ACT Short Mix reagents (Bioline USA Inc., Taunton, MA, USA). The pre-amplified samples were diluted five-fold with RNase- and DNase-free H 2 O.…”

“…In the non Myd88-dependent pathway, TLR4 triggers the activation of the TRIF-related adaptor molecule (TRAM), followed by the activation of interferon regulatory factor 3, which induces the production of IFN-γ. In the later inflammatory response process, TRAM can also activate NF-κB, inducing inflammatory factor expression and activation (Brempelis et al, 2017 ; Yan et al, 2017 ).…”
