Central Role for Interleukin-4 in Regulating Nitric Oxide-Mediated Inhibition of T-Cell Proliferation and Gamma Interferon Production in Schistosomiasis

Abstract: Interleukin-4 (IL-4) plays an important role in several areas of T-cell biology, including the development of Th2 responses (25) and blocking Th1 effector mechanisms mediated by gamma interferon (IFN-␥), such as inducible nitric oxide synthase (iNOS) induction (1,10,28,43,50). Further, IL-4 can inhibit NO production by upregulating arginase expression, thereby providing an alternative pathway for metabolism of L-arginine, the precursor for NO (34).The outcome of schistosome infection in IL-4 Ϫ/Ϫ animals is dif… Show more

“…iNOS, which produces nitric oxide from L-arginin, is induced during the immune response by microbial products and/or cytokines and is also involved in the anti-microbial effector mechanism of macrophages (10) . One of the T-helper 1 effector mechanisms mediated by IFN-g is iNOS induction, and IL-4 plays an important role as its regulator (11) . High numbers of iNOS (+) cells have been shown in tumour-bearing mice (DMH group), suggesting an increase in nitric oxide production by these cells.…”

“…Although the exact mechanism of IL-4 action in this system remains to be elucidated, a possible explanation for the ad- For example, IL-4 may inhibit macrophage-dendritic cell antiHistoplasma function by decreasing iNOS production or stimulating production of other cytokines that inhibit protection (26). Future experiments will examine the role of IL-4 on protective cellular functions.…”

“…19 Because IL-4 and IL-13 use the IL-4Ra chain for cell activation and are known regulators of NOS-2 expression, 20 we determined whether the absence of IL-4/IL-13 signaling affected NO production in our system. Although similar levels of NO were detected in splenocyte cultures from WT and IL-4Ra À/À mice 8 d.p.i., the absence of IL-4Ra led to a greater production of NO by splenocytes from mice 4 d.p.i.…”
