Central Nervous System Inflammation and Neurological Disease in Transgenic Mice Expressing the CC Chemokine CCL21 in Oligodendrocytes

Abstract: To study the biological role of the chemokine ligands CCL19 and CCL21, we generated transgenic mice expressing either gene in oligodendrocytes of the CNS. While all transgenic mice expressing CCL19 in the CNS developed normally, most (18 of 26) of the CCL21 founder mice developed a neurological disease that was characterized by loss of landing reflex, tremor, and ataxia. These neurological signs were observed as early as postnatal day 9 and were associated with weight loss and death during the first 4 wk of li… Show more

“…We detected CCL21 immunoreactivity in thyroid follicular cells, within the colloid material and in the thyroid parenchyma (Fig. 1B), suggesting that follicular cells secrete CCL21, as observed in other transgenic experiments (30). No immunoreactivity for CCL21 was observed in the control thyroids (Fig.…”

“…CCL21 and CXCL13 have been shown to induce formation of lymphoid structures when expressed in the pancreas (27)(28)(29). However, this property does not seem to be general, as expression of CCL21 in brain and skin of transgenic mice fails to induce development of such structures (30). Here, we directly test the hypothesis that the chemokine CCL21 may be directly implicated in regulating influx of lymphocytes into the thyroid.…”

“…Similarly, the expression of the murine CC chemokine JE in thymus, brain (39) Here we extend these observations by showing that both forms of CCL21 (Leu or Ser) can induce the formation of lymph node-like structures in the pancreas. Furthermore, we suggest a tissue-specific requirement for the biological activity of these molecules in the pancreas, because the expression of CCL21 in skin or brain (51) does not induce the same phenomenon.…”

“…CXCR3-deficiency was associated with reduced microglial activity and reduced loss of secondary neurons in hippocampal formation in an entorhinal cortex lesion model (57). Furthermore, ectopic CCL21 expression in oligodendrocytes induced a massive brain inflammation that killed the animals within 3 d after onset of the expression (55).…”

“…CCL21 and its receptor in the ischemic CNS, CXCR3, were previously found to be induced in ischemic mouse brain (54)(55)(56). CXCR3-deficiency was associated with reduced microglial activity and reduced loss of secondary neurons in hippocampal formation in an entorhinal cortex lesion model (57).…”
