Centaurin β1 Down-regulates Nucleotide-binding Oligomerization Domains 1- and 2-dependent NF-κB Activation

Abstract: Centaurin ␤1 (CENTB1), a GTPase-activating protein, is a member of the ADP-ribosylation factor family encoded by a gene located on the short arm of human chromosome 17. A yeast two-hybrid screen first suggested a direct interaction between CENTB1 and NOD2. Co-immunoprecipitation experiments confirmed direct interaction between CENTB1 and NOD2 and demonstrated similar interaction between CENTB1 and NOD1. We also demonstrate that endogenous CENTB1 interacts with endogenous NOD2 and NOD1 in SW480 and HT-29 intest… Show more

“…NOD2 also sensitizes epithelial cells to recognize H. pylori in vitro (19). NOD1 activity is regulated by its interacting proteins such as cIAP1/2 (26), SGT1 (27), CENTB (28), and Hsp90 (29). Multiple proteins, including TAK1 (30), , Ipaf-1 (31), Erbin (32), and CENTB1 (28) interact with NOD2 and regulate NOD2-mediated NF-κB activation.…”

“…The cell polarity protein Erbin, the GTPase-activating protein Centaurin-b1 (CENTB1) and the angioassociated migratory cell protein (AAMP) bind to Nod2 and negatively regulate Nod2-dependent NF-kB activation while the GRIM-19 protein that also associates with Nod2 appears to positively modulate Nod2 signalling [25,26,[45][46][47]. Since Erbin and CENTB1 expression is upregulated after Nod2 activation, both proteins could mediate a negative feedback.…”

“…Through the recognition of MDP, secretion of alpha-defensins is stimulated for protection against microbial invasion. In CD patients, a diminished expression of alpha-defensins has been found in those carriers with NOD2 mutation [20,21] . It is known that through this recognition of bacterial products the nuclear factor kappa B (NFκB) activation is regulated [10,22] .…”
