In woody crop plants, the oligosaccharide components of the cell wall are essential for important traits such as bioenergy content, growth, and structural wood properties. UDP-glucuronate decarboxylase (UXS) is a key enzyme in the synthesis of UDP-xylose for the formation of xylans during cell wall biosynthesis. Here, we isolated a multigene family of seven members (PtUXS1-7) encoding UXS from Populus tomentosa, the first investigation of UXSs in a tree species. Analysis of gene structure and phylogeny showed that the PtUXS family could be divided into three groups (PtUXS1/4, PtUXS2/5, and PtUXS3/6/7), consistent with the tissue-specific expression patterns of each PtUXS. We further evaluated the functional consequences of nucleotide polymorphisms in PtUXS1. In total, 243 single-nucleotide polymorphisms (SNPs) were identified, with a high frequency of SNPs (1/18 bp) and nucleotide diversity (πT = 0.01033, θw = 0.01280). Linkage disequilibrium (LD) analysis showed that LD did not extend over the entire gene (r 2<0.1, P<0.001, within 700 bp). SNP- and haplotype-based association analysis showed that nine SNPs (Q <0.10) and 12 haplotypes (P<0.05) were significantly associated with growth and wood property traits in the association population (426 individuals), with 2.70% to 12.37% of the phenotypic variation explained. Four significant single-marker associations (Q <0.10) were validated in a linkage mapping population of 1200 individuals. Also, RNA transcript accumulation varies among genotypic classes of SNP10 was further confirmed in the association population. This is the first comprehensive study of the UXS gene family in woody plants, and lays the foundation for genetic improvements of wood properties and growth in trees using genetic engineering or marker-assisted breeding.

Summary• Wood is an important biological resource which contributes to nutrient and hydrology cycles through ecosystems, and provides structural support at the plant level. Thousands of genes are involved in wood development, yet their effects on phenotype are not well understood. We have exploited the low genomic linkage disequilibrium (LD) and abundant phenotypic variation of forest trees to explore allelic diversity underlying wood traits in an association study.• Candidate gene allelic diversity was modelled against quantitative variation to identify SNPs influencing wood properties, growth and disease resistance across three populations of Corymbia citriodora subsp. variegata, a forest tree of eastern Australia.• Nine single nucleotide polymorphism (SNP) associations from six genes were identified in a discovery population (833 individuals). Associations were subsequently tested in two smaller populations (130-160 individuals), 'validating' our findings in three cases for actin 7 (ACT7) and COP1 interacting protein 7 (CIP7).• The results imply a functional role for these genes in mediating wood chemical composition and growth, respectively. A flip in the effect of ACT7 on pulp yield between populations suggests gene by environment interactions are at play. Existing evidence of gene function lends strength to the observed associations, and in the case of CIP7 supports a role in cortical photosynthesis.

ResumoA produção volumétrica tem sido prioridade nos programas de melhoramento florestal. No entanto, é necessário que a qualidade da madeira seja considerada, para que o fornecimento da matéria prima permita aumentar a eficiência e o rendimento industrial. Dessa forma, o objetivo deste trabalho foi estimar parâ-metros genéticos, correlações genéticas e ganhos com a seleção para caracteres de crescimento (diâme-tro), espessura de casca e densidade básica da madeira de Eucalyptus urophylla. Para tanto, utilizou-se discos de madeira retirados no DAP e metade da altura comercial, provenientes de árvores desbastadas em um teste de progênies de polinização aberta com 33 genótipos e duas procedências (Rio Claro-SP e Três Lagoas-MS), localizado em Selvíria-MS. Os parâmetros genéticos foram obtidos com base no procedimento REML/BLUP. O melhor desempenho para crescimento foi verificado para Rio Claro. Não foram detectadas diferenças genéticas para densidade da madeira entre as procedências. A procedência de Três Lagoas apresentou coeficiente de variação genética > 14% (diâmetro) e forte controle genético (herdabilidade > 0,6 e acurácia > 78%). Para qualidade da madeira, Rio Claro sobressaiu-se nas estimativas de herdabilidade (> 0,6 -DBM e ECD), alcançando acurácia superior a 79%. Foram encontrados resultados contrastantes para as correlações genéticas entre as duas procedências. Para a obtenção de materiais genéticos com maior densidade da madeira, a seleção para DBM é vantajosa. Palavras-chave: melhoramento florestal, qualidade da madeira, REML/BLUP. AbstractVolumetric production has been a priority in forest breeding programs. However, it is necessary to consider wood quality for the raw material supply that increases efficiency and yield. Thus, the aim of this study was to estimate genetic parameters, genitic correlations and selection gains for growth traits (diameter), bark thickness and basic density of wood in progênies of Eucalyptus urophylla. For this, we used wood discs removed at DHB and half the commercial height, from trees thinned in an open pollination progeny with 33 genotypes and two provenances (Rio Claro-SP and Três Lagoas-MS), located in Selvíria-MS. Estimates of genetic parameters were obtained using the REML/BLUP method. The best performance for growth was found in the Rio Claro provenance. Genetic differences in wood density between the provenances were not detected. The provenance of Três Lagoas showed a coefficient of genetic variation > 14% (diameter) and strong genetic control (heritability > 0.6 and accuracy > 78%). For wood quality, Rio Claro stood out in the estimates of heritability (> 0.6 -DBM and ECD), achieving a higher accuracy of 79%. Contrasting results were found for the genetic correlation between the two sources. For obtaining genetic materials with higher density wood, the selection for DBM is advantageous.
