Cellular toxicology and mechanism of the response to silver-based nanoparticle exposure in Ewing’s sarcoma cells

Abstract: Ewing’s sarcoma is the most aggressive connective tissue tumor, mainly affecting children and adolescents; the 5 year survival rate is only 50%. Current treatments have poor effectiveness, and more efficient treatments are being sought. Silver-based nanoparticles, such as silver chloride nanoparticles (AgCl-NPs) and silver/silver chloride (Ag/AgCl-NPs) nanoparticles, can be biologically produced and can release Ag+ ions into solution; however, their antitumor activity has been minimally investigated. The aim o… Show more

“…Cell research. Ag + induced mitochondrial apoptosis in human Chang liver cells, Ewing's sarcoma A673 cells, A549 lung cancer cells, human airway smooth muscle, human breast SKBR3 cancer cells, histiocytic lymphoma U-937 cells, human promyelocytic HL-60 cells, human keratinocytes, CHO-K line cells, human bronchial epithelial BEAS-2B cells, human skin fibroblasts, and mouse AML12 hepatocytes [89][90][91][92][93][94][95][96][97][98][99][100][101][102]. The apoptosis manifested alongside decreased cell viability, caspase 3 and 7 activation, pro-apoptotic genes (Bax and Bcl-2) expression, increased metallothionein expression, ATP depletion, and DNA fragmentation.…”

“…They include endothelial, perivascular lympho-endothelial, pure hematologic, mesenchymal/fibroblastic, neural crest derivatives, or stem/progenitor cells. The ultrastructural investigation of tissue specimens of ES/PNET reveals polygonal cells with slightly irregular nuclei, finely dispersed chromatin, and prominent nucleoli (14)(15)(16). Nucleoli are small-to-large and often open (nucleolonemas).…”
