Cellular Responses Required for Oxidative Stress Tolerance of the Necrotrophic Fungus Alternaria alternata, Causal Agent of Pear Black Spot

Abstract: To establish successful infections in host plants, pathogenic fungi must sense and respond to an array of stresses, such as oxidative stress. In this study, we systematically analyzed the effects of 30 mM H2O2 treatment on reactive oxygen species (ROS) metabolism in Alternaria alternata. Results showed that 30 mM H2O2 treatment lead to increased O2− generation rate and H2O2 content, and simultaneously, increased the activities and transcript levels of NADPH oxidase (NOX). The activities and gene expression lev… Show more

“…During the early stage of infection, A. alternata conidia adhere to the surface of pear fruit, where conidial spores gradually germinate into germ tubes [ 8 ]. A. alternata releases the host-specific toxins AK-I and AK-II, which block the host defense response [ 9 ]. A. alternata also secretes cell-wall-degrading enzymes during infection that enable the pathogen to penetrate and infect the host tissue [ 7 , 10 ].…”

“…Noxs genes are believe to regulate the ROS signals that are generated [ 14 ]. Past studies found that ROS are generated and accumulated at the infected site to prevent pathogen spread during pathogen infection [ 26 , 27 ]. To resist pathogen infection, the host initiated ETI, during which H 2 O 2 rapidly accumulated in and around the injection site when F. oxysporum infected the bitter gourd plants.…”

“…The assays of SOD, CAT, and POD activities were based on the methods previously described by Zhang et al (2022) and Wang et al (2021) [17,18]. In total, 0.2 g of frozen mycelium was ground in 5 mL of precooled phosphate buffer (50 mM and pH 7.8) and centrifuged at 4 • C and 9900× g for 20 min, and the supernatant was the crude enzyme of SOD and CAT.…”
