Cellular Remodeling During Plant Virus Infection

Abstract: This review focuses on the extensive membrane and organelle rearrangements that have been observed in plant cells infected with RNA viruses. The modifications generally involve the formation of spherules, vesicles, and/or multivesicular bodies associated with various organelles such as the endoplasmic reticulum and peroxisomes. These virus-induced organelles house the viral RNA replication complex and are known as virus factories or viroplasms. Membrane and organelle alterations are attributed to the action of… Show more

“…These virus-host interactions are often complex and difficult to understand mechanistically, but for some of them considerable progress has been made in recent years (Ahlquist et al, 2003;Boevink and Oparka, 2005;Benitez-Alfonso et al, 2010;Harries et al, 2010;Ishibashi et al, 2010;Laliberté and Sanfaçon, 2010;Verchot-Lubicz et al, 2010;Le Gall et al, 2011;Nagy and Pogany, 2011;Niehl and Heinlein, 2011;Schoelz et al, 2011). Progress also has been made toward understanding virus-host interactions involved in the plant immune response (Kang et al, 2005;Padmanabhan and Dinesh-Kumar, 2010;Cournoyer and DineshKumar, 2011).…”

“…Replication of all known eukaryotic positive-strand RNA viruses occurs in replication complexes formed on host membranes (6)(7)(8)(9)(10), which contain viral genomic RNAs, replication proteins, and host factors (11). To avoid elicitation of host defense systems triggered by viral double-stranded RNA (12), the activity of eukaryotic positive-strand RNA virus RNA-dependent RNA polymerases (RdRps) must be strictly regulated so that they synthesize negative-strand RNA only in the replication complexes that are sequestered from the cytoplasm.…”

“…Through the years, numerous studies have focused on the characterization of viral replication sites within the cell, as well as how plant virus movement proteins (MPs) modify the plasmodesmata to facilitate cell-tocell movement (for review, see Benitez-Alfonso et al, 2010;Laliberté and Sanfaçon, 2010;Niehl and Heinlein, 2011;Ueki and Citovsky, 2011;Verchot, 2012). It is accepted that plant virus replication is associated with host membranes, and at some point, the viral genomic nucleic acid must be transferred from the site of replication in the cell to the plasmodesmata.…”
