Cellular Populations Isolated from Newborn Mouse Skin Including Mesenchymal Stem Cells

Abstract: We developed protocols for isolation and characterization of mesenchymal progenitors from murine dermis. Our protocols are part of a more general isolation procedure starting with neonatal murine skin, which has been described in detail by U. Lichti and coauthors (Nat Protoc 3(5):799-810, 2008). We list Lichti's procedures in an abbreviated form as part of this methods section. Our methods to isolate mesenchymal stem cells are presented as a continuous workflow of isolation and characterization, including flow… Show more

“…Similar to NSCs, isolation of CSCs from tumor tissues, including thyroid cancers, have been performed by different approaches, including antigen-directed FACS sorting, side population and Aldefluor assay [ 52 , 68 ]. Although in vitro culture and maintenance of CSCs is difficult, the continuous development of new technologies has allowed the CSC culture as cell floating spheroids in non-adherent plates or 3D culture in reconstituted basal membranes (Matrigel) [ 69 , 70 ]. An important feature of CSCs, at various limiting dilution, is the ability to reproduce the original tumors in vivo.…”

“…Existing evidence suggests that skin might contain an MSC-like cell population ( Kimlin and Virador, 2013 ; Toma et al, 2001 ; Vaculik et al, 2012 ; Viswanathan et al, 2019 ). However, their biological properties and contribution to hematopoiesis and leukemia remain unexplored.…”

“…Existing evidence suggests that skin might contain an MSC-like cell population. [16][17][18][19] However, their biological properties and contribution to hematopoiesis and leukemia remain unexplored. Knowledge of this would be essential for understanding the pathology of extramedullary leukemia in the skin, and would thus provide new insights into identifying new potential therapeutic targets that may be translated into new treatment strategies for AML patients with leukemia cutis.…”
