Cellular Localization of Anti-DNP-PLL and Anticonveyor Albumin Antibodies in Genetic Nonresponder Guinea Pigs Immunized With DNP-PLL Albumin Complexes

Abstract: About 30-40% of random-bred Hartley (I) and all strain 2 (2) guinea pigs recognize poly-L-lysine (PLL) I and hapten-PLL conjugates as antigens. These "responder" animals develop delayed hypersensitivity to the conjugated polymer and produce specific antihapten antibodies when immunized with hapten-PLL conjugates. The ability to respond to these antigens was shown to be controlled by a dominant autosomal gene (2, 3). Hapten-PLL conjugates which are complete antigens only for responder animals can behave as hapt… Show more

“…The results of the present work are in good agreement with those of other studies [1,2,4,5,11,21] and support the concept of the by Immunocytes as Detected by Immunoenzymc Techniques 169 narrow specificity of the antibody synthesized by a single immunocyte and indicate that an immunologically competent cell, once engaged in the synthesis of a specific antibody, produces antibody of only this specificity. In the studies referred to above, about 3% mixed cells were noted.…”

“…Moreover, technologies by then available used a combination of immunofluorescence and historadi ography to identify the specificity of antibodies produced by individual plasma cells in animals immunized with hapten-protein conjugates. With Ira Green and Pierre Vassalli, we were delighted to find that the antibody producing cells of animals immunized with DNP-BSA or DNP-BGG produced antibodies specific for either the hapten or the carrier molecule, but never reactive with both, in spite of the presence of both determinants on the immunizing antigen (38,39). At about the same time Makela came to the identical conclusion by a different approach (40).…”
