Cellular immune mechanisms in myosin-induced myocarditis

Abstract: Cardiac myosin-induced myocarditis proved to be a valuable virus-free murine model with which to investigate autoimmunological mechanisms in inflammatory heart disease. The disease was shown to be T cell-mediated. In this contribution the functional role of CD4 and CD8 molecules and the conditions that are required to make the cardiac tissue susceptible to an autoimmune attack are discussed.

“…By day 15, most animals have cleared the virus. However, in certain genetically predisposed mice, antibodies are produced against one of the several myocytic components: the mitochondrial adenosine diphosphate/adenosine triphosphate carrier protein [8], myosin or laminin heavy chains [9], -adrenoceptors [10], or cardiac sarcoplasmic reticulum calcium adenosine triphosphatase [11]. Thus, a chronic inflammatory process is established in the absence of viral material.…”

“…The major target antigen for the autoimmune attack in mice is the cardiac a-myosin heavy chain (myhca). Myhca-induced autoimmune myocarditis in BALB/c mice is a T helper cell-mediated disease [9,10]. In DBA/2 mice, however, myhca-specific antibodies alone suffice to elicit myocardial injury if myhca is deposited on cardiomyocytes [11,12].…”
