Cellular and body scale morphology of <i>Heterocapsa huensis</i> sp. nov. (Peridiniales, Dinophyceae) found in Hue, Vietnam

One of the most common marine dinophytes is a species known as Heterocapsa triquetra. When Stein introduced the taxon Heterocapsa, he formally based the type species H. triquetra on the basionym Glenodinium triquetrum. The latter was described by Ehrenberg and is most likely a species of Kryptoperidinium. In addition to that currently unresolved nomenclatural situation, the thecal plate composition of H. triquetra sensu Stein (1883) was controversial in the past. To clarify the debate, we collected material and established the strain UTKG7 from the Baltic Sea off Kiel (Germany, the same locality as Stein had studied), which was investigated using light and electron microscopy, and whose systematic position was inferred using molecular phylogenetics. The small motile cells (18-26 μm in length) had a biconical through fusiform shape and typically were characterized by a short asymmetrically shaped, horn-like protuberance at the antapex. A large spherical nucleus was located in the episome, whereas a single pyrenoid laid in the lower cingular plane. The predominant plate pattern was identified as apical pore complex (Po, cp?, X), 4', 2a, 6'', 6c, 5s, 5''', 2''''. The triradiate body scales were 254-306 nm in diameter, had 6 ridges radiating from a central spine, 9 peripheral and 3 radiating spines, and 12 peripheral bars as well as a central depression in the basal plate. Our work provides a clarification of morphological characters and a new, validly published name for this important but yet formally undescribed species of Heterocapsa: H. steinii sp. nov.

RESUMOSão apresentadas informações sobre a composição florística e a distribuição espacial e temporal das espécies de cianobactérias e microalgas no Canal do Piraquê, situado na parte nordeste da Lagoa Rodrigo de Freitas, cidade do Rio de Janeiro, com base em 30 amostras quali-quantitativas coletadas nos meses de março, abril, junho, julho, outubro e dezembro de 2007 e em janeiro, fevereiro e março de 2008 na subsuperfície no ponto central do canal. Complementa-se a análise com uma comparação entre a composição florística do canal e do corpo central da lagoa. Foram identificados no canal 86 táxons (68 novos registros para o sistema Lagoa Rodrigo de Freitas) distribuídos principalmente em diatomáceas (39), clorofíceas (19) e cianobactérias (13). Nitzschia e Desmodesmus apresentaram o maior número de espécies. Pseudanabaena catenata, Synechocystis salina, Heterocapsa pygmaea e Thalassiosira weissflogii foram os táxons mais frequentes (70% das amostras). Houve predomínio de espécies limnobiontes (44%), enquanto táxons marinhos eurihalinos e limnobiontes eurihalinos contribuíram em igual proporção (20% cada). Espécies tipicamente salobras apresentaram menor contribuição (16%). Quando comparado com o corpo principal da lagoa, o canal mostrou predominância de Pseudanabaena spp. durante todo o período estudado, enquanto Pseudanabaena spp. e Synechocystis salina foram as espécies mais abundantes na lagoa nos períodos de estiagem e chuva, respectivamente. Em contrapartida, Heterosigma akashiwo, Cyclotella chocktawhatcheeana, Monoraphidium contortum e Monoraphidium irregulare ocorreram exclusivamente no canal. As diferenças na distribuição espacial e temporal das microalgas entre os dois compartimentos devem-se à ocorrência de florações pontuais, formadas por espécies típicas do canal. No corpo principal da lagoa, as populações de microalgas mostraram uma distribuição temporal menos variável, ao longo de todo o período de estudo. Characteristics important in identification of Heterocapsa species (i.e., thecal plate pattern, body scale structure, and shape and position of the nucleus and pyrenoid) are practically identical in the dinoflagellate investigated here and in Heterocapsa arctica T. Horig. described from the Canadian Arctic. Analysis of internal transcribed spacer (ITS) sequences confirms that the two dinoflagellates are very closely related; however, there is a clear difference in their size and shape. Our experiments show that the low‐salinity Baltic Sea brackish water does not reduce the size of the marine H. arctica to match that of the Baltic Sea morphotype. On the basis of these dissimilarities in general morphology and its geographic isolation in the Baltic Sea, we consider our material sufficiently differentiated from the typical H. arctica to warrant the status of a new subspecies, H. arctica subsp. frigida subsp. nov. Being of a distinct cell shape, the occurrence of subsp. frigida has been recorded in Algaline phytoplankton monitoring data collected since 1993. Although it has never been responsible for high biomass blooms, it commonly occurs in spring in the Northern Baltic Proper and in the western Gulf of Finland, when the water temperatures are <5°C.
