The largest energy consumer in the cell is the ribosome biogenesis whose aberrancy elicits various diseases in humans. It has been recently revealed that p53 induction, along with cell cycle arrest, is related with abnormal ribosome biogenesis, but the exact mechanism still remains unknown. In this study, we have found that aberrant ribosome biogenesis activates two parallel cellular pathways, c-Myc and ASK1/p38, which result in p53 induction and G1 arrest. The c-Myc stabilizes p53 by rpL11-mediated HDM2 inhibition, and ASK1/p38 activates p53 by phosphorylation on serine 15 and 33. Our studies demonstrate the relationship between these two pathways and p53 induction. The changes caused by impaired ribosomal stress, such as p53 induction and G1 arrest, were completely disappeared by inhibition of either pathway. These findings suggest a monitoring mechanism of c-Myc and ASK1/p38 against abnormal ribosome biogenesis through controlling the stability and activity of p53 protein.

Chromophobe renal cell carcinoma (RCC) is a form of renal cancer that may be confused with other eosinophilic renal tumors, including oncocytoma, type 2 papillary RCC, and clear-cell RCC with eosinophilic features. There are currently no robust markers to distinguish these neoplasms. Chromophobe RCC and renal oncocytoma are presumably derived from the distal nephron. FXYD2 is a distal tubule regulator of the trimeric Na þ / K þ -transporting ATPase that is enriched in kidney tissue. In this study, we investigated the expression of FXYD2 in normal human kidney, 27 chromophobe RCCs, 30 oncocytomas, 15 clear-cell RCCs, and 11 papillary RCCs. Immunohistochemical staining for FXYD2 showed diffuse, strong immunoreactivity in the basolateral membrane of distal tubules of normal human kidney. Ninety-six percent (26/27) of chromophobe RCCs were immunoreactive for FXYD2 in a distinctly membranous pattern. Twenty-five of these tumors showed at least focal 2 þ staining. In contrast, only 17% (5/30) of renal oncocytomas, 11% (2/15) of clear-cell RCCs, and 0% (0/11) of papillary RCCs displayed FXYD2 immunoreactivity. None of these cases showed Z2 þ FXYD2 staining. A subset of cases was confirmed as oncocytoma or chromophobe RCC using cytokeratin 7, colloidal iron, and interphase fluorescence in situ hybridization analysis of chromosomes 1, 2, 6, 10, and 17. Among this subset, 100% (7/7) of chromophobe RCCs were FXYD2 positive, whereas 17% (2/12) of oncocytomas were stained with FXYD2. The oncocytomas that stained with FXYD2 did so in a weak (1 þ ), patchy manner. In contrast, chromophobe RCCs showed Z2 þ staining in 86% (6/7) of these tumors. For comparison, this subset was also stained for kidney-specific cadherin (Ksp-cadherin). Ksp-cadherin showed positive staining in 100% (7/7) of chromophobe RCCs and 33% (4/12) of oncocytomas. This is the first report demonstrating the potential utility of FXYD2 immunohistochemistry in the diagnosis of chromophobe RCC.

Biogenèse des ribosomes et érythropoïèse : un lien inattenduLa biogenèse des ribosomes est l'activité métabolique la plus consommatrice d'énergie dans une cellule qui prolifère. La synthèse des 4 ARN et des 79 protéines ribosomiques qui composent les deux sous-unités ribosomiques mobilise l'essentiel de l'activité transcriptionnelle, les ARN messagers (ARNm) codant les protéines ribosomiques comptant pour la moitié des ARNm cellulaires. Par ailleurs, la production des ribosomes est étroitement connectée à la régulation du cycle cellulaire [1]. Malgré son rôle clé, peu de pathologies ont été liées à la mutation de gènes impliqués dans ce processus. Récemment, plusieurs maladies génétiques rares et un syndrome hématologique acquis ont été associés à la mutation de gènes codant une protéine ribosomique ou l'un des 300 facteurs requis pour la maturation des pré-ribosomes (particules précurseurs des sous-unités ribosomiques) [2]. De manière surprenante, ces pathologies sont presque toutes caractérisées par une insuffisance médullaire (Tableau I). Cette relation inattendue entre la biogenèse des ribosomes, un processus ubiquitaire, et le fonctionnement du tissu hématopoïé-tique est particulière-ment bien caractérisée dans le cas de l'anémie de Diamond-Blackfan, la première maladie génétique associée à la mutation de gènes codant des protéines ribosomiques. L'anémie de Diamond-BlackfanL'anémie de Diamond-Blackfan ou ADB (DiamondBlackfan anemia) est une maladie rare (5 à 7 cas par million de naissances vivantes) qui résulte d'un déficit sévère de la prolifération et de la différencia-tion des précurseurs érythroïdes. La moelle osseuse présente un nombre très réduit d'érythroblastes (moins de 5 % des cellules nucléées), alors que les autres lignées hématopoïétiques se différencient normalement. L'anémie apparaît dans les semaines qui suivent la naissance et est associée chez 40 % des patients à > L'anémie de Diamond-Blackfan (ADB) est une érythroblastopénie (absence ou déficit sévère des érythroblastes dans la moelle osseuse) congénitale associée à des mutations hétéro-zygotes dans des gènes codant des protéines ribosomiques. Des études récentes indiquent que ces mutations ont pour effet premier d'affecter la synthèse des ribosomes. L'explication du lien physiopathologique inattendu entre ce processus ubiquitaire et une maladie affectant particulièrement l'érythropoïèse pourrait en partie se trouver dans la notion émergente de « stress ribosomique », une voie d'arrêt du cycle cellulaire activée en réponse à un défaut de production des ribosomes. L'ADB est devenue un paradigme pour étudier le nombre croissant des maladies liées à la mutation de gènes codant des protéines entrant en jeu dans la biogenèse des ribosomes. <
