It is interesting that the mannoprotein we detected was metabolically labeled with L-[35S]rnethionine, since all determinations of amino acid composition of mannoprotein isolated by chemical methods from cell walls of C. albicans or S. cerevisiae have noted the absence of sulfur-containing amino acids (7,11,12). The reasons for this are unknown, but perhaps such amino acids are destroyed or lost by the extraction procedures used.…”

“…In this respect no differences in the major cytoplasmic proteins of any of the yeast or mycelial phase extracts have been identified (Manning & Mitchell, 1980). In addition, the overall wall composition of the two cellular forms has been shown to be similar (Chattaway et al, 1968), but the relative amounts of alkali-soluble and alkali-insoluble glucans (Sullivan et al, 1983) as well as the glycopeptides extracted with ethylenediamine (Kolarova et al, 1973) have been reported to change during germ-tube formation. Elorza et al (1983) showed that walls of yeast cells contain more mannan than those of the mycelial phase, suggesting that the incorporation of specific mannoproteins might be responsible for the different wall architecture.…”

“…The mannan and/3-glucan are complexed with proteins, which in the yeast phase are present as water-soluble, and water-insoluble glycopeptides, both forms composed of mannan peptides and glucan peptides (84). The ratio protein/carbohydrate of both types of water-soluble glycopeptides was higher in serotype A than in serotype B. Milder methods of extraction resulted in isolation of a series of three complex molecules, all of them with antigenic properties: a) peptidoglucomannan with a glucose: mannose ratio of 2:3, and an aminoacid content of 7.3%; b) a soluble mannoglucan with 11% mannose.…”
