Cell Volume to Aid Analysis and Technique of Somatic Cell Counts in Milk

Abstract: In conjunction with a Coulter Counter, somatic cells in milk were sized by electronic analysis. Quarter milk from cows with mastitis had a cell volume peak with a modal cell volume of 102 mu3 while milk from healthy quarters had no peak. Bulk milks with a peak had higher cell counts than milks where there was no peak. Dimensions of peak from bulk milks were the same as from quarter milks. Modal cell volume and cell count varied after milks were fixed with varying concentrations and types of fixative. The conc… Show more

“…A Coulter Channelyzer C1000 was interfaced to the Coulter Counter producing a distribution of the volumes of the cells counted for each milk sample. Details of the Channelyzer were given by Sheldrake et al (1977). The Channelyzer was adjusted to record all particles with a volume between 5 fi 3 and 180 /i 3 .…”

“…In samples from mastitic quarters the majority of cells had a cell volume of 50 to 150 /im 3 , with a mode of 102 fim 3 (Sheldrake et al 1977) which in the Model TA is covered by ch 6 and 7. Because of the weighting of channels, a substantial elevation in ch 7 above ch 6 indicates a peak of cells in ch 6 and 7.…”
