Cell type-dependent alterations of binding of synthetic blood group antigen-related oligosaccharides in lung cancer

Abstract: Blood group antigen-related oligosaccharides have been implicated in growth regulation, cell mobility control and adhesion; we are therefore interested in the localization of receptors for these oligosaccharides in tumour cells. Labelled neoglycoconjugates that carry synthetic sugar structures are suitable tools to determine: whether such binding sites are present in human lung cancer; whether structural alterations of the glycoligand part will affect extent of binding; and whether cell type-associated alterat… Show more

“…A number of OS, such as LacNAc, T ββ , GalNAcβ1-4GlcNAc, asialoGM1 and blood group A are known to be potent ligands for cellular lectins such as galectins [26]. Indeed, MDCK exhibited significant level of binding to LacNAc, asialoGM1, GalNAcβ1-4GlcNAc, TF, blood group A (A tri , A tetra ) and Fucα1-2Galβ1-3GlcNAc (Le d ) oligosaccharides as shown in Figure 1.…”

“…Statistical analysis was performed with chi-square test and the t-test. The biotinylated fucan was synthesized and ligandohistochemistry including the adequate specificity controls was performed, as described previously (Zeng et al, 1992;Kayser et al, 1994a). Histochemical reaction was classified to be positive, if all or clusters of pulmonary epithelial cells displayed a dark brown colour, and that of concomitant negative controls no detectable staining intensity.…”

“…Experimental details have been given elsewhere. 26,39 Additionally, gel filtration on Biogel P-10 was performed to remove any aggregates. The lectin was judged to be homogeneous by SDS-PAGE analysis.…”
