Cell Surface Biliverdin Reductase Mediates Biliverdin-induced Anti-inflammatory Effects via Phosphatidylinositol 3-Kinase and Akt

Abstract: Biliverdin reductase A (BVR) catalyzes the reduction of biliverdin (BV) to bilirubin (BR) in all cells. Others and we haveshown that biliverdin is a potent anti-inflammatory molecule, however, the mechanism by which BV exerts its protective effects is unclear. We describe and elucidate a novel finding demonstrating that BVR is expressed on the external plasma membrane of macrophages (and other cells) where it quickly converts BV to BR. The enzymatic conversion of BV to BR on the surface by BVR initiates a sign… Show more

“…The significance of hBVR phosphorylation by PKC␦ can be viewed in the context of its role in the cellular defense mechanisms against free radicals. As noted above, activation of hBVR can significantly influence those cellular functions that extend beyond its role in the cellular defense mechanisms (5,6,12,14,25,42,67).…”

“…BMDMs from mice were harvested and differentiated as previously described (68). For each experiment, cells were seeded at a density of 1 to 2 million per 60 mm plate.…”

“…BM cells were isolated and cultured in the medium containing SCF (100 ng/ml), IL-6 (10 ng/ml), and IL-3 (6 ng/ml) (PeproTech) for 24 hours. Retroviral microadapted shRNA was prepared as described (68). Cells were transduced 12 hours with retroviruses in medium containing Polybrene (8 μg/ml, Sigma-Aldrich).…”

“…PC3 cells were transiently transfected with siRNA against STMN1 or scramble siRNA using Lipofectamine 2000 (Invitrogen) as previously described. 38 Twenty four hours post-transfection, cells were split to 96 well plates and treated with CITs for 72h. To confirm successful knockdown, we measured STMN1 levels by immunoblotting.…”
