Cell Patterning Method on a Clinically Ubiquitous Culture Dish Using Acoustic Pressure Generated From Resonance Vibration of a Disk-Shaped Ultrasonic Transducer

Abstract: Cell patterning methods have been previously reported for cell culture. However, these methods use inclusions or devices that are not used in general cell culture and that might affect cell functionality. Here we report a cell patterning method that can be conducted on a general cell culture dish without any inclusions by employing a resonance vibration of a disk-shaped ultrasonic transducer located under the dish. A resonance vibration with a single nodal circle patterned C2C12 myoblasts into a circular shape… Show more

“…Every proposed new structure oriented to the operation target has to do all the work from beginning to end. [69,70], Lu et al [71], and Imashiro et al [72,73]. (a) Very small micropillers are machined equidistantly along two symmetrical spiral lines.…”

“…This effect can be observed in the vibration of the drumhead. Imashiro et al [72,73] proposed a manipulator where a vibrating glass Using silicon substrate vibration to control particle motion is another method. The vibration generated or propagated through the substrate can induce the ARF.…”

“…However, manual steps present a risk of contamination and result in variable culture efficiency and stability between trials and depending on the technician [6]. In the interest of standardizing these processes, several techniques have been developed, such as cell patterning, detaching, collecting, and harvesting [7]- [12]. Cell counting technologies, which are key to realizing stable and effective cell culture, have been developed, but they require dedicated devices or labor-intensive processes [13], [14].…”

“…Fluid flow through a microfluidic device applies shear stress to cells cultured in the device [14]. Vibrational stimulation is gaining attention because it is easy to apply in the ubiquitous cell culture dish format and is highly biocompatibile [15]. The method using vibration stimulation has the advantage that the culture environment is not changed, compared with the conventional migration regulation method represented by using chemical stimulation.…”
